当归多糖胶原缓释微球复合新型生物活性多孔支架的制备及其细胞相容性的研究

【摘要】:目的制备并研究当归多糖明胶缓释微球复合硫酸钙/冻干骨支架的细胞相容性。方法支架用煅烧挂浆法制备,分别在原支架和表面修饰后的支架上复合培养第三代成骨细胞,利用倒置显微镜观察两种支架上的成骨细胞数目及形态等,利用扫描电镜观察复合支架和成骨细胞复合支架培养时的支架表面;评估方法为细胞蛋白质和细胞黏附率测定法。结果在复合成骨细胞培养的各个时期,实验组的蛋白质含量较对照组含量多,并且实验组的细胞黏附率也高于对照组(P0.05)。提示新型支架较原支架更有利于细胞生长,并且电镜下观察到支架孔隙中也有细胞生长。结论经过当归多糖修饰后的支架较原支架的细胞相容性高。
[Abstract]:Objective to prepare and study the cytocompatibility of Angelica polysaccharide gelatin sustained-release microspheres combined with calcium sulfate / freeze-dried bone scaffold. Methods the scaffolds were prepared by calcination-suspension method. The third generation osteoblasts were co-cultured on the original scaffold and the surface modified scaffold respectively. The number and morphology of osteoblasts on the scaffolds were observed by inverted microscope. The surface of composite scaffold and osteoblast composite scaffold was observed by scanning electron microscope (SEM). Results the protein content of the experimental group was higher than that of the control group, and the cell adhesion rate of the experimental group was higher than that of the control group. The results indicated that the new scaffold was more favorable to cell growth than the original scaffold, and the cell growth was observed in the pore of the scaffold under electron microscope. Conclusion the scaffold modified by Angelica polysaccharide has higher cytocompatibility than the original scaffold.
