胶原支架联合CBD-VEGF在大鼠全横断脊髓损伤中的应用

【摘要】:目的:本研究通过建立SD大鼠脊髓第九胸椎(T9)全切损伤模型,评价胶原支架(CS)联合胶原结合区-血管内皮生长因子(CBD-VEGF)移植对脊髓损伤的修复作用。方法:80只健康雄性6周龄SD大鼠,体重为215±15g,随机分成4组:1.假手术组;2.对照组;3.CS移植组;4.CS/CBD-VEGF移植组。术后对各组大鼠每周进行一次BBB评分,并在术后第4周、10周和12周末施行组织学检查及免疫组化等检查,在10周、12周末行双侧腓肠肌测重及Masson染色。结果:根据排除准则,所有实验对象手术成功,各实验组无大鼠因脊髓横断不完全而淘汰,共计80只大鼠纳入研究。在术后4周末CS/CBD-VEGF组的生存率明显高于对照组与CS组,但总体趋势无统计学意义。术后7周,CS/CBD-VEGF组与CS组的双下肢BBB评分逐渐高于对照组(p≤0.05),CS组与对照组间评分差异则至10周后逐步出现(p≤0.05)。4周、10周及12周的免疫组化分析可见:CS/CBD-VEGF组anti-NF、anti-NSE及anti-CD34染色密度明显比CS组与对照组高,anti-GFAP染色密度较后两者低,说明脊髓损伤处神经纤维及血管再生情况较CS组及对照组好,且有较少的瘢痕组织形成。12周末腓肠肌重量分析可见:CS/CBD-VEGF组双侧腓肠肌重量明显比CS组与对照组高(p≤0.05),同组双下肢间无明显差异(p0.05);Masson三色染色腓肠肌密度及粗细较CS组及对照组高(p≤0.05),表明损伤双下肢肌肉萎缩情况较后两组轻。结论:大鼠脊髓全横断损伤后,CS/CBD-VEGF的植入能够有效改善损伤处局部微环境,抑制瘢痕组织的形成,促进血管及神经纤维的再生与双下肢功能的恢复。
[Abstract]:Objective: to establish a model of total spinal cord injury in Sprague-Dawley rats (Sprague-Dawley) 9th thoracic spine (T9). Objective: to evaluate the effect of collagen scaffold combined with vascular endothelial growth factor (CBD-VEGF) graft on the repair of spinal cord injury (sci). The body weight was 215 卤15g, and was randomly divided into 4 groups: 1: 1. Sham-operated group; 2. Control group; 3. CS transplantation group; 4. CSP / CBD-VEGF transplantation group. The rats in each group were evaluated with BBB once a week after operation, and the histological and immunohistochemical examinations were performed at the 4th week and the 12th week after operation. Results: according to the rule of exclusion, all the subjects were successfully operated, and no rats in each experimental group were eliminated because of incomplete transection of spinal cord. A total of 80 rats were included in the study. The survival rate of CS/CBD-VEGF group was significantly higher than that of control group and CS group at the end of 4 weeks, but the overall trend was not statistically significant.
