脐血干细胞移植联合电针对脊髓损伤修复作用的研究

【摘要】:目的:观察移植脐血干细胞(human umbilical cord blood stem cell, hUCBSC)联合电针治疗(Electro-acupuncture, EA)对脊髓全横断大鼠的神经功能的恢复作用。方法:本次实验采用原代离心培养的方式收集培养脐血干细胞,调整细胞浓度为1.0×107/μL,移植备用。挑选雌性健康成年SD大鼠72只,重量220-240 g,随机共分为4组:损伤模型组(SCI)、脐血干细胞移植组(hUCBSC)、督脉电针治疗组(EA)、脐血干细胞移植联合督脉电针治疗组(hUCBSC+EA),每组18只。各组分脊髓横断损伤后7d、14d、28d三个监测时间点。制作大鼠脊髓全横断模型。术后,hUCBSC笼给予一次干细胞移植,细胞浓度为1.0×107/μJ L。EA笼和EA+hUCBSC笼每天电针治疗一次,电针治疗的时间和频率设置为30min、5hz,直至取材。首先用Basso-Beaie-Bresnehan scale(BBB)运动评分评价大鼠SCI后神经修情况,然后运用PowerLab数据记录系统检测不同时间点大鼠运动诱发电位的传导,最后行电镜观察,分析各组治疗后脊髓修复情况在形态结构方面的改变。结果:1、大鼠脊髓全横断后的,各组大鼠随着时间的延长,后肢活动均呈阶梯式的恢复且联合组与另外三组存在差异(P0.05),联合组动物肢体功能恢复显著。2、电镜观察后发现时间越长,髓鞘的形态结构恢复越明显,空泡越少,肿胀越轻,且联合治疗组的新生髓鞘最多,形态规整。3、运动诱发电位检测显示,电针治疗组检测结果与脐血干细胞移植组差异不明显(P0.05),且联合组结果与另外三组存在差异(P0.05)。结论:脊髓损伤(SCI)后,脐血干细胞能分化迁移并作用于脊髓横断部位;电针刺激能够作用于神经损伤部位,脐血干细胞与电针刺激联合治疗能促进神经髓鞘再生,使大鼠脊髓损伤后的运动功能明显恢复。
[Abstract]:Objective: to observe the effect of umbilical cord blood stem cell (hUCBSC) combined with electroacupuncture on the recovery of neural function in rats with spinal cord transection. Methods: umbilical cord blood stem cells were collected and cultured by primary centrifugation. The cell concentration was adjusted to 1.0 脳 107 / 渭 L. 72 adult female SD rats were selected. Weighing 220-240 g, randomly divided into 4 groups: injury model group, cord blood stem cell transplantation group, umbilical cord blood stem cell transplantation group, du vein electroacupuncture group, umbilical cord blood stem cell transplantation combined with du vein electroacupuncture treatment group, hUCBSC EAA group, 18 rats in each group. The rat spinal cord transection model was made at a time point of monitoring. After operation, the hUCBSC cage was given a stem cell transplantation. The cell concentration was 1.0 脳 10 7 / 渭 J L.EA cage and EA hUCBSC cage once a day. The time and frequency of electroacupuncture treatment was set at 30 min or 5 h, until the material was taken. The nerve repair after SCI was evaluated by Basso-Beaie-Bresnehan scale-BBB) score. Then PowerLab data recording system was used to detect the conduction of motor evoked potential in rats at different time points. Finally, electron microscopy was used to analyze the morphological and structural changes of spinal cord repair after treatment in each group. With the extension of time, the hind limb activity of each group showed a stepwise recovery, and there was a difference between the combined group and the other three groups (P 0.05). The recovery of limb function in the combined group was significant. The longer the time was, the more obvious the morphological structure of the myelin sheath was in the combined group, the longer the time was, the more obvious the recovery of the myelin structure was. The fewer vacuoles, the lighter the swelling, and the combined treatment group had the most new myelin sheath, shape. 3, motor evoked potential test showed, The results of electroacupuncture treatment group were not significantly different from those of cord blood stem cell transplantation group (P 0.05), and the results of combined group were different from those of the other three groups (P 0.05). Conclusion: after spinal cord injury (SCI), cord blood stem cells can differentiate and migrate and act on spinal cord transection. The combined treatment of umbilical cord blood stem cells and electroacupuncture stimulation can promote the regeneration of nerve myelin sheath and recover the motor function after spinal cord injury in rats.
