白花丹素对骨肉瘤细胞增殖、凋亡的影响及其机制

【摘要】:目的探讨白花丹素对骨肉瘤细胞MG-63(简称骨肉瘤细胞)增殖、凋亡的影响,并探讨其可能的机制。方法将对数生长期的骨肉瘤细胞随机分为四组,5μmol/L组、10μmol/L组和20μmol/L组分别加入含5、10和20μmol/L白花丹素的培养基,正常对照组加入含0.1%DMSO的完全培养基。培养24 h,采用CCK-8法检测细胞增殖抑制率,Hoechst33342染色后计算细胞凋亡率,流式细胞术检测活性氧(ROS)水平,Western blotting法检测Bcl-2、Bax蛋白相对表达量。结果对照组、5μmol/L组、10μmol/L组、20μmol/L组细胞增殖抑制率和凋亡率均逐渐升高,组间两两比较P均0.05。对照组和5μmol/L组、10μmol/L组、20μmol/L组ROS水平、Bax蛋白相对表达量均逐渐升高,Bcl-2蛋白相对表达量逐渐降低,组间比较P均0.05。结论白花丹素能够抑制骨肉瘤细胞增殖、促进其凋亡,并呈浓度依赖性;升高细胞内ROS水平、激活线粒体途径可能是其作用机制。
[Abstract]:Objective to investigate the effect of Baihuadan on proliferation and apoptosis of osteosarcoma cell line MG-63 (osteosarcoma cell line). Methods Osteosarcoma cells in logarithmic phase were randomly divided into four groups: 5 渭 mol/L group, 10 渭 mol/L group and 20 渭 mol/L group. The normal control group was cultured on a complete medium containing 0.1% DMSO for 24 h. The inhibition rate of cell proliferation was detected by CCK-8 assay and the apoptosis rate was calculated by Hoechst33342 staining. The level of reactive oxygen species (Ros) was detected by flow cytometry and the relative expression of Bcl-2P Bax protein was detected by Western blotting. Results the proliferation inhibition rate and apoptosis rate of control group (5 渭 mol/L), 10 渭 mol/L group (10 渭 mol/L) and 20 渭 mol/L group increased gradually. The relative expression of ROS protein in 10 渭 mol/L group and 10 渭 mol/L group of 5 渭 mol/L group increased gradually and decreased gradually in 20 渭 mol/L group, while the relative expression of Bcl-2 protein decreased gradually in the control group and 5 渭 mol/L group (P 0.05). Conclusion Bax protein can inhibit the proliferation of osteosarcoma cells. Promoting its apoptosis in a concentration-dependent manner, increasing the level of ROS and activating the mitochondrial pathway may be the mechanism of its action.
