大鼠反复轻度脑创伤后外周血中T细胞亚群的变化

【摘要】：目的:探讨大鼠反复轻度脑创伤(rm TBI)后外周血中T细胞亚群的变化及其意义。方法:选择SD大鼠,采用控制性脑皮质撞击法(CCI)制作rm TBI大鼠模型,随机分为假手术对照组以及损伤后1、2、4、6周组共5组,每组8只,通过流式细胞仪检测各组大鼠1、2、4、6周时外周血中CD3~+、CD4~+、CD8~+细胞百分率。结果:CD3~+T细胞百分率在1、2周均有显著上升,在4、6周均有显著下降(P0.05)。CD4~+T细胞百分率在伤后持续下降(P0.05);CD8~+T细胞百分率在伤后1~4周逐渐上升(P0.05)。结论:rm TBI模型大鼠在伤后存在免疫功能紊乱,这为rm TBI伤后存在外周免疫抑制提供了新的实验证据,其慢性期外周免疫抑制可能对疾病的预后产生重要影响。
[Abstract]:Objective: to investigate the changes and significance of T cell subsets in peripheral blood of rats after repeated mild brain trauma (RM TBI). Methods: SD rats were selected and the rat model of RM TBI was established by controlled cerebral cortex impact. Eight rats in each group were randomly divided into 5 groups: sham operation control group and 6-week group with 8 rats in each group. The percentage of CD3 ~ + CD4 ~ + CD8 ~ cells in peripheral blood of rats in each group was detected by flow cytometry at 6 weeks after injury. Results the percentage of CD3 ~ T cells increased significantly at 1 ~ 2 weeks after injury. There was a significant decrease in the percentage of P0.05U. CD4- T cells at the 6th week after injury. The percentage of CD8- T cells increased gradually at the 1st and 4th week after injury. Conclusion there is a disturbance of immune function in the rat model of TBI at 4 weeks after injury. This provides new experimental evidence for the existence of peripheral immunosuppression after rm TBI injury, and its chronic peripheral immunosuppression may have an important effect on the prognosis of the disease.
