多途径靶向给药系统在骨肉瘤治疗中的研究进展

【摘要】:骨肉瘤是好发于儿童及青少年的恶性骨肿瘤,由于骨组织硬度大、渗透性差和生理生化过程的特殊性,抗肿瘤药物在骨组织很难达到有效的药物浓度。选择增加药物剂量不仅提高了药物毒副反应的发生率,也给患者在治疗过程中带来心理及经济上的压力。实验研究发现了多种途径能使抗肿瘤药物通过自我导向或在外界环境诱导下选择性地与肿瘤组织结合,其中包括通过骨诱导剂(双膦酸盐类)偶联化疗药物、纳米药物载体封装化疗药物以及小分子核糖核酸结合化疗药物等方式。本文就目前国内外研究抗肿瘤药物通过靶向给药系统治疗骨肉瘤的实验成果、理论机制及治疗方向作综述。
[Abstract]:Osteosarcoma is a malignant bone tumor that occurs in children and adolescents. Due to its high bone hardness, poor permeability and particularity of physiological and biochemical processes, osteosarcoma is a malignant tumor in children and adolescents. Antitumor drugs are difficult to achieve effective drug concentration in bone tissue. Choosing to increase the dosage of drugs not only increases the incidence of adverse drug reactions, It also brings psychological and economic pressure to patients during the course of treatment. Experimental studies have found that many ways can enable anti-tumor drugs to selectively bind to tumor tissue through self-directed or induced by external environment. These include coupling chemotherapy drugs with bone inducers (bisphosphonates), Nano-drug carriers encapsulate chemotherapeutic drugs and small molecule ribonucleic acid combined with chemotherapeutic drugs. The theoretical mechanism and therapeutic direction are reviewed.
