医用臭氧对腰椎间盘突出症患者髓核组织的结构以及NO和IL-6表达的影响

【摘要】:目的:观察不同浓度医用臭氧对腰椎间盘突出症患者髓核组织的结构改变以及NO和IL-6表达的影响。方法:经椎间孔镜取出的腰椎间盘突出症患者髓核16例,每例随机分为5组:对照组(C组)不施加任何干预,氧气组(O2组)、医用臭氧20μg/ml组(0320组)、医用臭氧40μg/ml组(0340组)和医用臭氧60μg/ml组(0360组)分别用O2、20μg/mlO 40μg/mlO3和60μg/mlO3干预20分钟。体外培养72h后,HE染色观察髓核组织结构变化,分别采用硝酸酶还原法和酶联免疫吸附法(ELISA)检测培养液NO和IL-6含量,实时荧光定量PCR检测iNOS mRNA和IL-6 mRNA表达。结果:1.髓核组织结构改变:与对照组比较,O320组和O340组髓核组织的基质成分改变和纤维沉积不明显,0360组基质成分减少,排列紊乱,胶原纤维增生。2.NO和IL-6表达变化:与C组比较,O2组NO和IL-6的表达差异无统计学意义(P0.05);与C组和02组比较,0320组和0340组NO和IL-6表达降低(P0.01),而0360组NO和IL-6表达升高(P0.05)。0320组和0340组之间NO和IL-6的表达差异无统计学意义(P0.05)。3. iNOS mRNA和IL-6 mRNA表达变化:与C组比较,02组iNOS和IL-6mRNA的表达都无明显差异(P0.05)。0320组和0340组iNOS mRNA表达明显下降(P0.05),而0360组iNOS mRNA表达升高(P0.05)。同样,与C组比较,0320组和0340组IL-6 mRNA表达也显著下降(P0.05),而O360组IL-6 mRNA表达也明显上升(P0.05)。结论:医用臭氧具有降解病变髓核组织基质成分和促纤维沉积作用。20μg/ml和40μg/ml医用臭氧可抑制腰椎间盘突出症患者髓核iNOS mRNA和IL-6 mRNA的表达,减少NO合成,降低炎性反应。60μg/ml医用臭氧增加NO和iNOS mRNA以及IL-6和IL-6 mRNA的表达。
[Abstract]:Objective: to observe the effects of different concentrations of medical ozone on the structural changes of nucleus pulposus and the expression of no and IL-6 in the nucleus pulposus of patients with lumbar disc herniation. Methods: sixteen patients with lumbar disc herniation were treated by intervertebral foramen. Each case was randomly divided into 5 groups: control group C) without any intervention, O _ 2 / O _ 2 group, medical ozone 20 渭 g / ml group 0 320, medical ozone 40 渭 g / ml group 0 340) and medical ozone 60 渭 g / ml group 0 360) were treated with O _ 2O _ (20 渭 g / ml O _ (40) 渭 g / ml O _ (3) and 60 渭 g / ml O _ (3) for 20 minutes, respectively. After 72 hours of culture in vitro, the structure of nucleus pulposus was observed by HE staining. The contents of no and IL-6 in culture medium were detected by nitrate reduction method and enzyme linked immunosorbent assay (Elisa), respectively. Real time fluorescence quantitative PCR was used to detect the expression of iNOS mRNA and IL-6 mRNA. Results 1. Structural changes of nucleus pulposus: compared with the control group, the matrix composition and fiber deposition of nucleus pulposus in O320 group and O340 group were not obviously changed and the matrix components in group 0360 were decreased and disordered. 2. Changes of no and IL-6 expression: there was no significant difference in the expression of no and IL-6 between group C and group C, but the expression of no and IL-6 in group C and group 0340 decreased the expression of no and IL-6 in group C and group 02, while the expression of no and IL-6 in group 0360 increased the expression of no and IL-6 in group P 0.05.0320. There was no significant difference in the expression of no and IL-6 between groups 0 340 and 0 340. There was no significant difference in the expression of iNOS mRNA and IL-6mRNA between group 0 and group 0 340. There was no significant difference in the expression of iNOS and IL-6mRNA between group 2 and group C, but the expression of iNOS mRNA in group 0 340 and group 0 320 decreased significantly, while the expression of iNOS mRNA increased in group 0 360. Compared with group C, the expression of IL-6 mRNA in group C0320 and group 0340 also decreased significantly, while the expression of IL-6 mRNA in group O360 also increased significantly. Conclusion: medical ozone can degrade the matrix components of diseased nucleus pulposus and promote the deposition of fibers. 20 渭 g / ml and 40 渭 g / ml of medical ozone can be used to treat the lesion. To inhibit the expression of iNOS mRNA and IL-6 mRNA in nucleus pulposus of patients with lumbar disc herniation. The expression of no, iNOS mRNA, IL-6 and IL-6 mRNA was increased by reducing the synthesis of no and decreasing the inflammatory response. 60 渭 g / ml of medical ozone increased the expression of no, iNOS mRNA and IL-6 mRNA.
