条件培养基局部移植对大鼠脊髓损伤的修复作用

【摘要】:目的观察骨髓间充质干细胞的条件培养基(BMSC-CM)局部移植对脊髓损伤修复的作用。方法采用改良的Allen装置制作大鼠T9脊髓损伤模型。随机分为对照组(脊髓损伤后局部微量注射生理盐水10μl)和实验组(脊髓损伤后局部注射BMSC-CM 10μl),两组均采用BBB评分、斜板实验法观察大鼠运动功能恢复情况,苏木精-伊红(HE)染色观察脊髓损伤处血管生长及空洞体积的改变情况。结果BMSC-CM局部移植组的大鼠BBB后肢功能评分、斜板的倾斜角度明显高于对照组(P0.05),脊髓损伤处的空洞体积明显小于对照组,血管直径大于对照组。结论在彻底避免了BMSCs的直接和间接作用下用BMSC-CM能够减小脊髓损伤处的空洞体积和促进血管生长,从而促进脊髓损伤后神经功能的恢复。
[Abstract]:Objective to observe the effect of local transplantation of bone marrow mesenchymal stem cells (BMSCs) on the repair of spinal cord injury. Methods the model of T9 spinal cord injury in rats was made by modified Allen device. BBB score was used in both groups (BMSC-CM 10 渭 l after spinal cord injury) and experimental group (10 渭 l normal saline). The recovery of motor function and the changes of vascular growth and cavity volume in spinal cord injury were observed by hematoxylin and eosin (HEH) staining. Results the BBB hind limb function score of rats in BMSC-CM local transplantation group was observed. The inclined angle of the inclined plate was significantly higher than that of the control group (P 0.05), and the volume of the cavity in the spinal cord injury was obviously smaller than that in the control group. Conclusion under the direct and indirect action of BMSCs, BMSC-CM can reduce the volume of cavities and promote the growth of blood vessels in spinal cord injury, thus promoting the recovery of nerve function after spinal cord injury.
