丙泊酚后处理对大鼠肝缺血再灌注时肾组织细胞凋亡Bcl-2和Bax的影响

【摘要】:目的:研究丙泊酚后处理对大鼠肝脏缺血再灌注时肾脏细胞B淋巴细胞瘤/白血病-2(Bcl-2),Bcl相关X蛋白(Bax)的影响。方法:健康雄性SD大鼠54只,随机分成3组:假手术(S)组,缺血再灌注(IR)组,丙泊酚(P)组,(n=18)每组再根据再灌注时间(2h、4h、6h)的不同分成3个时相,每个时相为一个亚组,每个亚组各6只大鼠。分别为S2、S4、S6、IR2、IR4、IR6、P2、P4、P6亚组,共九个亚组。完成制作约70%肝缺血再灌注的模型,除S组之外,其余各组均为缺血45min后恢复灌注,S组、IR组缺血35min时以0.3ml·kg-1·min-1持续静脉泵注0.9%生理盐水,P组缺血35min时以0.3ml·kg-1·min-1持续泵注0.1%丙泊酚溶液。泵注时间均为10min,即3mg/kg。实验结束即刻取大鼠左肾下极,行组织切片和苏木精-伊红(HE)染色,光镜下观察。检测各样本大鼠血清中半胱氨酸蛋白酶抑制剂C(Cys-c)的浓度。使用免疫组化方法检测各组肾组织Bcl-2和Bax蛋白含量的表达,TUNEL法计算肾脏细胞的凋亡指数(AI)。结果:1、与S组相比,IR组和P组的Bcl-2、Bax表达增多(P0.05);与IR组相比,P组Bcl-2蛋白表达增多,Bax蛋白表达减少(P0.05)。2、与S组相比,IR组和P组的AI值均增高(P0.05);与IR组相比,P组的AI值减少(P0.05)。3、与S组相比,IR组和P组的Cys-c含量均增高(P0.05);与IR组相比,P组的Cys-c含量减少(P0.05)。4、光镜下IR组肾组织病理损伤变化比P组明显。结论:1、丙泊酚对肝缺血再灌注后不同时刻的肾脏损伤具有保护作用。2、这种保护机制可能与调控Bcl-2,Bax蛋白的表达有关。
[Abstract]:Objective: to study the effect of propofol postconditioning on renal B lymphocyte tumor / leukaemia Bcl-2Bcl related X protein Bax.Methods: 54 healthy male SD rats were randomly divided into 3 groups: sham operation group. Each group was divided into 3 time phases according to the reperfusion time of 2 h ~ 4 h ~ (6 h). Each phase was a subgroup with 6 rats in each subgroup. There were nine subgroups. About 70% liver ischemia-reperfusion model was made, except group S, The rest groups were treated with 0.3ml kg-1 min-1 continuous infusion of 0.1% propofol solution during 35min ischemia with 0.3ml kg-1 min-1 at the end of the experiment. The injection time was 10 min, that is, 3 mg / kg. The left subrenal pole of the rats was taken immediately after the end of the experiment, and the rest groups were injected with 0.3ml kg-1 min-1 continuously at the end of the experiment, and the rats were injected with 0.1% propofol solution at the end of the experiment. Tissue sections and hematoxylin-eosin (HEH) staining were performed. The concentration of cysteine protease inhibitor (Cy Cys-c) in serum of rats was detected under light microscope. The expressions of Bcl-2 and Bax protein in renal tissues were detected by immunohistochemical method. The apoptotic index of renal cells was calculated by Tunel method. Results compared with S group and P group, the expression of Bcl-2nBax protein increased and the expression of Bcl-2 protein decreased in P group compared with S group, and the AI value in P group and P group was higher than that in group S and P group (P 0.05), and the AI value in P group was lower than that in P group (P < 0.05), while that in P group was significantly higher than that in P group (P < 0.05), the expression of Bcl-2 protein in P group was higher than that in group S and P group (P < 0.05). Compared with group S, the content of Cys-c in IR and P groups was higher than that in group S, and the content of Cys-c in group P was lower than that in group IR. The pathological changes of renal tissue in IR group were more obvious than that in group P. Conclusion 1 and propofol have different effects on hepatic ischemia-reperfusion injury. This protective mechanism may be related to the regulation of the expression of Bcl-2P Bax protein.
