淫羊藿苷对大鼠肩袖损伤重建术后腱骨愈合的影响

【摘要】:目的:探索淫羊藿苷对大鼠肩袖损伤重建术后腱骨愈合的影响。方法:健康雄性Sprague Dawley(SD)大鼠(体重250-300g)64只,随机分为淫羊藿苷组(即ICA组,n=32)和对照组(n=32)。将冈上肌肌腱于肱骨大结节止点处切断后采用骨隧道法原位缝合,建立大鼠肩袖损伤重建模型。ICA组大鼠术后每天以125mg/kg淫羊藿苷灌胃,对照组中大鼠每日给予相同剂量的生理盐水灌胃。两组分别于术后2周、4周各处死一半大鼠(每组16只)并收集标本,通过Micro-CT扫描、血清生化、生物力学和组织学检测等手段评估淫羊藿苷对大鼠肩袖损伤重建术后腱骨愈合的影响。结果:血清生化检测提示ICA组术后2周、4周碱性磷酸酶及钙蛋白浓度显著高于对照组(P0.05)。Micro-CT扫描提示ICA组骨密度及骨体积分数(BV/TV)显著高于对照组(P0.05)。与对照组相比,ICA组腱骨交界检测到更多的软骨样细胞、成骨细胞、Collagen Ⅰ和VEGF阳性细胞。生物力学测定发现ICA组最大抗拉强度大于对照组(P0.05)。两组标本术后各时间点Collagen Ⅲ的表达水平无显著差异(P0.05)。结论:实验结果提示淫羊藿苷可促进腱骨交界区域血管及新骨形成并可增强肩袖重建术后力学性能,对大鼠肩袖损伤重建术后腱骨早期愈合有潜在促进作用。
[Abstract]:Objective: to investigate the effect of icariin on tendon healing after rotator cuff reconstruction in rats. Methods: healthy male Sprague Dawley SD rats (body weight 250-300g)64) were used in this study. Icariin group (ICA group) and control group (control group) were randomly divided into two groups. The supraspinatus muscle tendon was severed at the insertion point of the great tubercle of humerus and then sutured in situ with bone tunnel method to establish the rotator cuff injury reconstruction model of rats. ICA group rats were perfused with 125mg/kg icariin every day after operation. Rats in the control group were given the same dose of normal saline daily. Half of the rats (16 rats in each group) died 2 weeks and 4 weeks after operation, and the samples were collected, and the serum biochemistry was detected by Micro-CT scanning. Effects of icariin on tendon healing after rotator cuff reconstruction in rats were evaluated by biomechanical and histological methods. Results: serum biochemical examination showed that alkaline phosphatase and troponin concentrations in ICA group were significant at 2 weeks and 4 weeks after operation. Compared with control group, bone mineral density and bone volume fraction of ICA group were significantly higher than that of control group (P 0.05). More chondroid cells were detected in tendon bone junction of ICA group than those in control group. The maximum tensile strength of ICA group was higher than that of control group (P0.05).
