β磷酸三钙和α半水硫酸钙复合人工骨生物相容性及在脊柱融合模型中的应用

【摘要】:背景:β磷酸三钙和α半水硫酸钙备的人工复合骨具有多孔颗粒形态,生物相容性较高,用于脊柱融合模型中有助于融合率的提高,但该结论尚未得到证实。目的:观察β磷酸三钙/α半水硫酸钙复合人工骨的制备方法、生物相容性及在脊柱融合模型中的应用效果。方法:(1)将二水硫酸钙在定点的条件、温度下脱水制备α半水硫酸钙;取健康牛松质骨经脱细胞、脱脂后在特点的条件、温度下煅烧制备β磷酸三钙多孔颗粒,溶于无水乙醇中,混悬,烘干后制备β磷酸三钙和α半水硫酸钙复合人工骨。取购置的兔骨膜成骨细胞与复合材料共同培养,观察细胞形态、黏附能力及增殖活力;(2)取新西兰大白兔30只,建立双侧新西兰大白兔胸椎多节段后外侧脊柱融合模型,左侧植入β磷酸三钙和α半水硫酸钙复合人工骨,右侧植入自体骨进行对照,比较2组融合率变化。结果与结论:(1)细胞形态:相差显微镜观察可见,L929系细胞株培养3 d后在β磷酸三钙和α半水硫酸钙复合人工骨上贴壁数量相对较少;培养5 d后细胞在人工骨上贴壁相对密集;扫描电镜下β磷酸三钙和α半水硫酸钙复合人工骨表面存在许多结晶颗粒,表面吸附细胞相对较多;(2)β磷酸三钙和α半水硫酸钙复合人工骨植入后4周融合率,高于自体骨(P0.05);(3)自体骨植入后4周组织切片中骨侧小梁稀疏杂乱,可见新生的骨组织,无核的自体骨移植骨块占据主导地位;融合后8周自体骨移植骨周围新生骨组织进一步增多;β磷酸三钙和α半水硫酸钙复合人工骨植入后4周材料结构清晰可见,未见降解碎片,内部存在少许新生骨组织;植入后8周材料被新生骨组织包围,骨小梁增粗,人工骨开始降解;(4)结果证实:制备的β磷酸三钙和α半水硫酸钙复合人工骨用于脊柱融合模型中能获得较高的融合率。
[Abstract]:Background: the artificial composite bone prepared by 尾 tricalcium phosphate and 伪 -hemihydrate calcium sulfate has porous particle shape and high biocompatibility. It is helpful to improve the fusion rate in spinal fusion model, but this conclusion has not been confirmed.Objective: to observe the preparation method, biocompatibility and application effect of 尾 tricalcium phosphate / 伪 hemihydrate calcium sulfate composite artificial bone in spinal fusion model.Methods (1) calcium sulfate dihydrate was dehydrated at a fixed point to prepare 伪 -hemihydrate calcium sulfate, and healthy bovine cancellous bone was removed from cells, then calcined at temperature to prepare 尾 -tricalcium phosphate porous particles, which were dissolved in anhydrous ethanol.The composite artificial bone of 尾 -tricalcium phosphate and 伪 -hemihydrate calcium sulfate was prepared by suspension and drying.尾 tricalcium phosphate and 伪 -hemihydrate calcium sulfate composite artificial bone were implanted on the left side and autogenous bone was implanted on the right side to compare the fusion rate between the two groups.Results and conclusion: the cell morphology was observed by phase contrast microscope. After 3 days of culture, the number of adherent cells on 尾 tricalcium phosphate and 伪 -hemihydrate calcium sulfate composite artificial bone was relatively small, and the cell adhesion on artificial bone was relatively dense after 5 days of culture.4 weeks after autologous bone implantation, the bone trabeculae were sparse and disorganized. The new bone tissue was found, and the nucleless autologous bone grafts occupied the dominant position.8 weeks after implantation, the materials were surrounded by new bone tissue, the trabecular bone thickened, and the artificial bone began to degrade. The results showed that the 尾 -tricalcium phosphate and 伪 -hemihydrate calcium sulfate composite artificial bone could obtain high fusion rate in spinal fusion model.
