DON毒素与氟对骨软骨联合损伤机制的研究

【摘要】:目的观察DON毒素和氟对TGF-β2等4种骨生长发育相关指标的影响,探讨DON毒素联合元素氟致骨软骨的损伤机制。方法 28日龄Wistar大鼠40只,按照随机化方法分为空白组、氟(F)组、DON毒素组和"DON+F"组。F组和"DON+F"组饮用150 mg/L Na F溶液,空白组和DON毒素组饮用去离子水;各组的DON毒素染毒按0.2μg/g剂量每天灌胃,空白组和F组用等量蒸馏水灌胃。喂养5周后取血,ELISA法测定各细胞因子。结果血清中TGF-β2的平均含量"DON+F"组(55.47±5.07 pg/ml)DON组(38.88±7.99 pg/ml)F组(38.29±5.61 pg/ml)对照组(29.20±6.12 pg/ml);血清中BMP-2的平均含量"DON+F"组(15.24±0.66 ng/ml)F组(14.70±1.88 ng/ml)DON组(13.94±0.70 ng/ml)对照组(12.44±1.07 ng/ml);血清中Hyp的平均含量"DON+F"组(7.30±1.20μg/ml)DON组(7.38±1.65μg/ml)F组(8.35±1.51μg/ml)对照组(12.51±1.79μg/ml);血清中TNF-α的平均含量"DON+F"组(936.88±126.55 pg/ml)F组(860.09±117.45 pg/ml)DON组(706.52±73.72 pg/ml)对照组(579.54±144.53pg/ml)。结论 DON毒素和F存在一定的交互作用,可导致染毒大鼠血清TGF-β2等细胞因子的变化,进而影响骨软骨的生长发育,导致病理损伤。
[Abstract]:Objective to observe the effects of DON toxin and fluorine on four related indexes of bone growth and development, such as TGF- 尾 2, and to explore the mechanism of bone cartilage injury induced by DON toxin and fluoride. Methods Forty Wistar rats aged 28 days were randomly divided into blank group. Don toxin group, "DON F" group and "DON F" group drank 150 mg/L NaF solution, blank group and DON toxin group drank deionized water. The serum TGF- 尾 2 was measured by Elisa after 5 weeks of feeding. Results the average content of TGF- 尾 2 in serum of "DON F" group was 55.47 卤5.07 pg/ml)DON group (38.88 卤7.99 pg/ml)F) control group (29.20 卤6.12 PG / ml) and control group (29.20 卤6.12 PG / ml), and the average content of BMP-2 in serum was "DON F" in the control group (29.20 卤6.12 PG / ml). The average content of TNF- 伪 in the control group was 12.44 卤1.07 ng / ml, in the control group was 12.44 卤1.07 ng / ml, in the "DON F" group was 7.30 卤1.20 渭 g / ml, in the "DON F" group was 7.30 卤1.65 渭 g/ml)F group (8.35 卤1.51 渭 g / ml), in the control group was 12.51 卤1.79 渭 g / ml / ml, and in the DON F group was 936.88 卤126.55 pg/ml)F group (866.52 卤73.72 PG / ml). Conclusion there is a certain interaction between DON and F in the control group. It can cause the change of TGF- 尾 2 and other cytokines in the serum of exposed rats, and then affect the growth and development of bone cartilage and lead to pathological injury.
