雌激素对兔脊髓缺血再灌注损伤保护作用的机制研究

【摘要】:目的研究雌激素对脊髓缺血再灌注损伤保护作用的机制。方法按随机数表字法把成年的新西兰大白兔分为A~E 5组,每组9只。钳夹肾下腹主动脉20 min恢复再灌注;B组,接受与A组同样的麻醉和手术准备,但不行缺血/再灌注损伤;C组,再灌注开始时从兔耳的缘静脉注射200μg/kg雌激素;D组,再灌注开始时从兔耳的缘静脉注射400μg/kg雌激素;E组,再灌注开始时从兔耳的缘静脉注射800μg/kg雌激素。再灌注48 h后根据Tarlov法对下肢的神经功能进行评分,然后取脊髓组织苏木素-伊红染色法(HE)进行病理分析。结果 Tarlov评分中雌激素组的神经功能评分高于A组,与A组比较有统计学意义(P0.05)。组织病理表明脊髓的前角运动神经元出现凋亡,有显著坏死。A组脊髓的前角运动神经元计数少于雌激素组,两者比较差异有统计学意义(P0.05)。结论雌激素可明显改善兔脊髓缺血再灌注的下肢神经功能,使脊髓的前角正常运动神经元的数量增加,减小缺血再灌注的损伤。
[Abstract]:Objective to study the protective effect of estrogen on spinal cord ischemia reperfusion injury.Methods the adult New Zealand white rabbits were divided into five groups according to the random numeration method, 9 rabbits in each group.Group B received the same anesthesia and operation preparation as group A, but no ischemia / reperfusion injury. Group D received 200 渭 g/kg estrogen at the beginning of reperfusion.At the beginning of reperfusion, 400 渭 g/kg estrogen was injected into the marginal vein of rabbit ear and 800 渭 g/kg estrogen was injected into the vein of rabbit ear at the beginning of reperfusion.After 48 hours of reperfusion, the nerve function of lower extremity was assessed by Tarlov method, and then the histopathological analysis was performed by hematoxylin-eosin staining in spinal cord tissue.Results the neurological function score of estrogen group was higher than that of A group in Tarlov score, which was significantly higher than that in A group (P 0.05).Histopathology showed that the number of anterior horn motor neurons in group A was less than that in estrogen group, and there was significant difference between the two groups (P 0.05).Conclusion estrogen can obviously improve the nerve function of lower extremity after spinal cord ischemia reperfusion, increase the number of normal motor neurons in the anterior horn of spinal cord, and reduce the injury of ischemia reperfusion.
