柚皮苷对椎间盘退变的作用及其机制研究

【摘要】:目的:研究柚皮苷对体外椎间盘退变模型的作用,以及相应的作用机制。 方法:实验所需的大鼠髓核细胞来自于大鼠的尾椎椎间盘。利用IL-1β作用于髓核细胞建立椎间盘退变的细胞模型。用不同浓度的柚皮苷作用于处理后的髓核细胞,对比它对基质金属蛋白酶表达的影响,以及对于转录因子NF-κB及其通路的影响。 结果:柚皮苷能够明显降低由IL-1β刺激髓核细胞的基质金属蛋白酶的表达,并能够通过提高IκB-α的表达水平来抑制NF-κB通路的活化。 结论:柚皮苷是一种有效的抗炎和抗分解代谢药物,通过抑制NF-κB信号通路在髓核细胞中发挥作用,可以被研发为一种治疗椎间盘退变的有效药物。
[Abstract]:Aim: to study the effect of naringin on the model of disc degeneration in vitro and its mechanism. Methods: the rat nucleus pulposus cells were obtained from the rat caudal disc. The cell model of disc degeneration was established by the action of IL-1 尾 on nucleus pulposus cells. The effects of naringin at different concentrations on the expression of matrix metalloproteinase and the transcription factor NF- 魏 B and its pathway were compared. Results: naringin could significantly decrease the expression of matrix metalloproteinases stimulated by IL-1 尾 and inhibit the activation of NF- 魏 B pathway by increasing the expression of I 魏 B- 伪. Conclusion: naringin is an effective anti-inflammatory and anti-catabolism drug. It can be developed as an effective drug for the treatment of intervertebral disc degeneration by inhibiting NF- 魏 B signaling pathway in nucleus pulposus cells.
