大鼠皮肤的保存与撕脱伤模型冷藏皮延迟植皮的实验研究

【摘要】:目的对比研究冷藏皮和反植皮的成活率及冷藏皮和液氮冷冻皮肤的病理学变化。方法分析冷藏皮延迟植皮的成活率及病理学变化情况,并对冷藏皮和液氮冷冻皮进行病理形态学和细胞凋亡率观察。结果动物实验研究表明,即时回植组与冷藏皮24 h、48 h组成活率无统计学差异(P0.05);冷藏皮72 h、120 h、168 h回植组成活率明显低于即时回植组(P0.05),提示冷藏皮回植黄金时间为48 h以内。大鼠皮肤细胞空泡变性率比较,冷藏皮在第1,3,5,10天的空泡变性率均高于即时回植组(P0.05),但第1天的空泡变性率接近即时回植组,而第3,5,10天的空泡变性率与即时回植组差距较大。液氮组第1,3,5,10天的空泡变性率也高于即时回植组(P0.05),但是空泡变性率均较接近即时回植组,组间绝对数差距也较冷藏组的组间差距小。大鼠皮肤细胞凋亡率比较,冷藏皮在第1,3,5,10天的凋亡率均高于即时回植组(P0.05),但第1、3天的凋亡率无统计学差异(P0.05),而与第5、10天的凋亡率差距较大。液氮组的第1,3,5,10天的凋亡率明显高于即时回植组(P0.05),凋亡率也明显高于冷藏组。结论皮肤冷藏48 h以内为回植的安全期限,液氮冷藏皮肤,细胞变性率低,但是冻存和解冻过程可能导致细胞凋亡率增加。
[Abstract]:Objective to study the survival rate of cold storage skin and anti-graft skin and the pathological changes of refrigerated skin and liquid nitrogen frozen skin. Methods the survival rate and pathological changes of frozen skin and liquid nitrogen frozen skin were analyzed, and the pathomorphology and apoptosis rate of frozen skin and liquid nitrogen frozen skin were observed. Results Animal experiments showed that there was no significant difference in survival rate between the immediate replanting group and the cold storage skin group at 24 h and 48 h, and the survival rate of the cold storage skin group was significantly lower than that of the immediate replanting group within 48 h. The survival rate of the cold storage skin group was significantly lower than that of the immediate replanting group within 48 h. Compared with the denaturation rate of skin cells, the denaturation rate of frozen skin on day 10 was higher than that of immediate replantation group (P 0.05), but the rate of denaturation of frozen skin was close to that of immediate replantation group on day 1, but the rate of denaturation of vacuolation on day 3 510 was larger than that of group B. The denaturation rate of vacuolation in liquid nitrogen group was also higher than that in immediate replantation group on the 10th day, but the denaturation rate of cavitation was similar to that of immediate replantation group, and the absolute difference between groups was also smaller than that in cold storage group. The apoptotic rate of skin cells in the cold storage skin group was higher than that in the immediate replantation group on the 10th day, but there was no significant difference in the apoptosis rate on the 1st day and the 3rd day, but there was a big difference between the cold storage skin and the 5th day. The apoptosis rate of liquid nitrogen group was significantly higher than that of immediate replantation group on the 10th day, and the apoptotic rate was significantly higher than that of cold storage group. Conclusion the safe time of skin replantation is within 48 h, and the denaturation rate of skin is low when liquid nitrogen is refrigerated, but the rate of apoptosis may be increased during freezing and thawing.
