拉喹莫德灌胃对大鼠腰椎间盘退变的治疗作用

【摘要】：目的观察拉喹莫德灌胃对大鼠腰椎间盘退变的治疗作用。方法将60只大鼠随机分为空白对照组(A组)、模型对照组(B组)、拉喹莫德组(C组)各20只。B、C组建立腰椎间盘退变模型,术后2周C组给予拉喹莫德按60 mg/(kg·d)灌胃,A、B组予以等体积的生理盐水灌胃,连续6周持续给药。第8周处死各组大鼠,手术取出L_(4~5)节段腰椎间盘中的髓核组织,采用光镜和电镜观察大鼠椎间盘髓核组织病理,采用Western blot法检测大鼠椎间盘髓核组织中HIF-2α。结果 C组较B组退变坏死细胞数量明显减少,脊索细胞数目较多仅少量细胞核遭到破坏,胶原纤维和周期性横纹大部分可见且有序。A、B、C组大鼠椎间盘髓核组织中HIF-2α相对表达量分别为115.6±12.5、150.3±18.2、127.2±18.7,两两相比,P均0.05。结论拉喹莫德灌胃对大鼠腰椎间盘退变具有治疗作用,这可能是通过减少髓核组织中HIF-2α的表达实现的。
[Abstract]:Objective to observe the therapeutic effect of laquimod on lumbar disc degeneration in rats. Methods Sixty rats were randomly divided into three groups: control group A (n = 20), model control group B (n = 20), laquimod group C (n = 20) and control group C (n = 20). Two weeks after operation, group C was given the same volume of normal saline by intragastric administration of laquimod for 60 mg/(kg d, and the drug was given continuously for 6 weeks. At the 8th week, the rats in each group were killed, and the nucleus pulposus tissue was removed from the lumbar intervertebral disc of L _ (4 / 5) segment by operation. The histopathology of the nucleus pulposus was observed by light microscope and electron microscope, and the HIF-2 伪 in the nucleus pulposus tissue of the rat disc was detected by Western blot method. Results the number of degenerative necrotic cells in group C was significantly lower than that in group B, and the number of notochord cells was more than that in group B. The relative expression of HIF-2 伪 was 115.6 卤12.5150.3 卤18.2127.2 卤18.7 in the nucleus pulposus of the intervertebral disc in most of the collagenous fibers and periodic transverse striae, and the relative expression of HIF-2 伪 in the nucleus pulposus was 115.6 卤12.5150.3 卤18.2127.2 卤18.7in the control group (P < 0.05). Conclusion Laquimod has therapeutic effect on the degeneration of lumbar intervertebral disc in rats, which may be achieved by reducing the expression of HIF-2 伪 in nucleus pulposus.
