辛伐他汀对去势大鼠椎体骨量及椎间盘退变的作用

【摘要】:目的辛伐他汀作为降脂类药物表现出一定的促进骨形成作用。文中旨在探讨辛伐他汀对卵巢切除大鼠椎体骨的骨量、微观结构以及椎间盘退变的作用。方法所有大鼠采用随机数字表法分为3组:假手术组,模型组、辛伐他汀组,每组10只。假手术组切开暴露卵巢,但不予切除;其余2组大鼠均行双侧卵巢切除术,并分别灌胃给予等渗盐水和辛伐他汀干预(10 mg/m L)溶于双蒸水制成混悬液,给药剂量5 mg/(kg·d)。取大鼠L3椎体行micro-CT分析其微观结构,取L5-6椎体,双能X线检测其骨密度后常规脱钙包埋,制备切片并行VG染色,经组织学观察和半定量评分评估L5-6椎间盘退变情况。结果与假手术组L5、L6、L5-6骨密度[(0.2933±0.011 0)、(0.290 7±0.015 0)、(0.286 0±0.011 5)g/cm2]比较,模型组[(0.242 3±0.008 1)、(0.239 5±0.006 1)、(0.238 0±0.005 9)g/cm2]降低,辛伐他汀组[(0.2598±0.0249)、(0.2572±0.0121)、(0.2528±0.0126)g/cm2]降低(P0.05);与模型组比较,辛伐他汀组L5、L6、L5-6骨密度升高(P0.05)。三维重建图显示,与假手术组比较,模型组和辛伐他汀组骨小梁稀疏,连结性较差。BV/TV、Tb.N降低,Tb.Sp升高(P0.05);辛伐他汀组Tb.Sp升高(P0.05)。椎间盘组织学观察及评分:假手术组脊索细胞较多、细胞外基质较丰富、纤维环排列整齐。模型组、辛伐他汀组相比于假手术组,脊索细胞数量减少并被软骨样细胞取代,有黏液样改变,纤维环与髓核边界模糊、排列紊乱。模型组、辛伐他汀组半定量评分[(4.35±0.90)分、(3.53±0.42)分]显著高于假手术组[(2.48±0.92)分],差异有统计学意义(P0.05),辛伐他汀与模型组比较差异无统计学意义(P0.05)。结论卵巢切除大鼠不仅椎体骨量丢失,椎间盘也有退变表现;辛伐他汀可以部分抑制卵巢切除大鼠腰椎骨量丢失,但对该模型椎间盘退变无显著影响。
[Abstract]:Objective simvastatin, as a lipid lowering drug, showed a certain effect on bone formation. The purpose of this study was to explore the effect of simvastatin on bone mass, microstructure and disc degeneration in ovariectomized rats. Methods all rats were divided into 3 groups by random digital table method: artificial hand group, model group, simvastatin group, 10 rats in each group. In the operation group, the ovaries were cut and exposed, but the other 2 groups of rats were treated with bilateral oophorectomy, and they were treated with isotonic saline and simvastatin (10 mg/m L) to dissolve in double water to make a suspension, 5 mg/ (kg. D). The microstructures of the L3 vertebral body of rats were analyzed by micro-CT, and L5-6 vertebral body was taken and the double energy X ray was used to detect the bone density. After the routine decalcification, the L5-6 disc degeneration was evaluated by histological observation and semi quantitative score. The results were compared with the L5, L6, L5-6 bone density of the sham operation group [(0.2933 + 0.0110), (0.2907 + 0.0150), (0.2860 + 0.0115) g/cm2], and the model group [0.2423 + 0.0081), (0.2395 + 0.0061), (0.2380 + 0.0059) g/cm 2] decreased, simvastatin Group [(0.2598 + 0.0249), (0.2572 + 0.0121), (0.2528 + 0.0126) g/cm2]) decreased (P0.05). Compared with the model group, L5, L6, L5-6 bone density increased (P0.05) in the simvastatin group. The three-dimensional reconstruction map showed that the bone trabecula in the model group and simvastatin group was sparse, the connective poor.BV/TV, Tb.N decreased, Tb.Sp increased (P0.05). The Tb.Sp of the simvastatin group increased (P0.05). The histological observation and score of the intervertebral disc: the sham operation group had more chord cells, more abundant extracellular matrix and neatly arrayed fibrous ring. The semi quantitative score of the model group and simvastatin Group [(4.35 + 0.90), (3.53 + 0.42)] was significantly higher than that of the sham operation group ((2.48 + 0.92)), the difference was statistically significant (P0.05). There was no significant difference between the simvastatin and the model group (P0.05). Simvastatin can partially inhibit lumbar bone loss in ovariectomized rats, but has no significant effect on intervertebral disc degeneration in this model.
