椎间盘退变患者髓核组织中HtrA1的表达及其与MMPs的关系

【摘要】：目的观察椎间盘退变患者髓核组织中高温必需因子A1(HtrA1)的表达及其与基质金属蛋白酶(MMPs)的关系。方法 78例接受椎间盘髓核切除患者,其中年轻脊柱骨折患者(改良Thompson分级Ⅰ级)10例,椎间盘退变患者68例(改良Thompson分级Ⅱ级22例、Ⅲ级24例、Ⅳ级22例)。采用real-time PCR和Western blotting法分别检测椎间盘髓核组织中的Htr A1及MMP-1、3、7、9、13 mRNA和蛋白,并采用Pearson法分析Htr A1和MMP-1、3、7、9、13的相关性。结果改良Thompson分级Ⅰ、Ⅱ、Ⅲ、Ⅳ级患者椎间盘髓核组织中HtrA1 mRNA分别为0.56±0.49、0.70±0.45、1.47±0.86、4.10±1.17,蛋白分别为0.16±0.09、0.52±0.07、1.21±0.26、3.10±0.17,两两相比P均0.05。不同改良Thompson分级患者椎间盘髓核组织中MMP-1、3、13的mRNA、蛋白表达两两相比P均0.05。HtrA1 mRNA与MMP-1、3、13 mRNA表达呈正相关(r~2分别为0.144 8、0.240 6、0.869 5,P均0.01)。结论椎间盘髓核组织中HtrA1的表达随椎间盘退变程度的升高而升高,且与MMP-1、3、13的表达有关。
