大鼠弥漫性轴索损伤后髓鞘碎片聚积和清除

【摘要】:目的:检测大鼠弥漫性轴索损伤后(diffuse axonal injury,DAI)髓鞘碎片在轴索损伤位点的堆积和清除。 方法:雄性SD大鼠25只,随机分为实验组20只和对照组5只。实验组大鼠采用Marmarou方法制作大鼠DAI模型,分别于伤后1天、7天、14天和28天各取5只,断头取脑,制作冰冻切片,并采用油红O染色检测损伤位点的髓鞘碎片。对照组5只进行假手术操作而不打击,并采用同样的方法处理脑组织。显微镜下观察各切片,计数油红O染色阳性颗粒的个数。 结果:对照组在皮层、海马及脑干内均未见明显RO染色阳性颗粒。实验组中,伤后1天,以上区域脑组织内仍未见明显阳性颗粒。在皮层组织内,伤后7天平均每200x图片内阳性颗粒的数目为48.00±16.70个;到14天时,增加至366.67±149.39个,与7天时相比具有显著性差异(p0.01);至28天时,阳性颗粒数目降至25.67±7.02个,与7天时数目基本相当(与28天组比,p0.01;与7天组比,p0.05)。在海马组织内,伤后7天平均每200x图片内阳性颗粒的数目为11.67±3.06个;至14天时为23.33±4.16个,较7天组显著增加(p0.01);至28天时,阳性颗粒基本消失,平均数目为2.00±1.00个(与7天组及14天组相比,均为p0.01)。在脑干组织内,伤后7天平均每200x图片内阳性颗粒的数目为51.67±6.81个;14天时为23.67±4.93个,较7天时显著降低(p0.01),28天时平均每200x图片内阳性颗粒的数目与14天时基本相当,为26.69±5.69个(与7天组比p0.01,与14天组比p0.05)。 结论:DAI后在大鼠新皮层、海马及脑干组织中存在着髓鞘碎片长时间的聚积,可能通过其继发性反应影响DAI的预后。
[Abstract]:Aim: to investigate the accumulation and clearance of diffuse axonal injuryDAI fragments at axonal injury sites after diffuse axonal injury in rats. Methods: 25 male SD rats were randomly divided into experimental group (n = 20) and control group (n = 5). The DAI model of rats in the experimental group was made by Marmarou method. 5 rats were taken from each group on the 1st day, the 7th day, the 14th day, and the 28th day, the head was cut off, and the frozen sections were made, and the myelin sheath fragments at the injury site were detected by oil red O staining. In the control group, 5 rats underwent sham operation without attack, and the brain tissue was treated with the same method. Each section was observed under microscope and the number of oil red O positive granules was counted. Results: no RO positive granules were found in the cortex, hippocampus and brain stem in the control group. In the experimental group, 1 day after injury, no obvious positive granules were found in the brain tissue of the above areas. In cortical tissue, the average number of positive particles per 200x image was 48.00 卤16.70 on the 7th day after injury, increased to 366.67 卤149.39 on the 14th day, which was significantly different from that of the 7th day, and decreased to 25.67 卤7.02 at 28 days. The number was about the same as that at 7 days (compared with 28 days group, P 0.01, and 7 days group, P 0. 05%, P 0. 05%, P 0. 05%, P 0. 05%). In hippocampal tissue, the average number of positive particles per 200x image was 11.67 卤3.06 on the 7th day after injury, 23.33 卤4.16 on the 14th day, which was significantly higher than that in the 7th day group, and the positive granules disappeared basically at 28 days. The average number was 2.00 卤1.00 (compared with 7-day group and 14-day group, p0.01). In brain stem tissue, the average number of positive particles per 200x image at 7 days after injury was 51.67 卤6.81 and 23.67 卤4.93 on day 14, which was significantly lower than that at day 7, and the number of positive particles per 200x image on day 28 was basically the same as that at day 14. It was 26.69 卤5.69 (p 0.01 vs 7 d and p 0.05 5) compared with 14 d group. Conclusion there is a long accumulation of myelin fragments in the neocortex, hippocampus and brain stem tissue of rats after the brain stem injury, which may influence the prognosis of DAI through its secondary reaction.
