急性脊髓损伤大鼠移植骨髓间充质干细胞后神经功能恢复及Nogo-A表达的影响

【摘要】:目的:探讨骨髓间充质干细胞BMSCs(Bone Marrow Mesenchymal Stem Cells)经大鼠尾静脉移植对大鼠急性脊髓损伤SCI(Spinal cord injury)后神经功能的恢复及对Nogo-A表达水平的影响。方法:采用全骨髓贴壁方法分离、培养、纯化骨髓间充质干细胞,大鼠急性脊髓损伤模型采用改良艾伦法建立。实验采用成年的雌性SD大鼠96只,随机性分成4个组,24只/组,分别是假手术组,SCI组,Vehicle组,BMSCs治疗组。骨髓间充质干细胞BMSCs移植前,用羧基荧光素二醋酸盐琥珀酰亚胺酯(carboxyfluorescein Succinimidyl Ester,CFSE)标记。BMSCs组于造模成功10分钟后,自大鼠尾静脉注射BMSCs悬液1ml(5×106个BMSCs);vehicle组则注射同等剂量的PBS。分别在术后第1天、第3天、第7天和第14天采用BBB神经行为学评分对神经行为功能进行评估,观察其神经行为功能恢复情况,采用免疫组化及RT-PCR法测定脊髓Nogo-A表达水平。结果:BMSCs大鼠移植BMSCs成功后,可以在脊髓损伤神经的周边组织区域中,观察到阳性CFSE染色细胞;BMSCs大鼠组损伤后第7天和第14天神经功能评估明显优于损伤对照组、溶剂对照组;大鼠BMSCs组术后第3天、第7天和第14天脊髓损伤区周边神经组织中Nogo-A表达比较损伤对照组和溶剂对照组,均有明显降低(P0.01)。.结论:急性脊髓损伤大鼠移植骨髓间充质干细胞,促进损伤后大鼠脊髓神经为功能的康复,部分机制与骨髓间充质干细胞抑制或下调脊髓组织中Nogo-的生成有关。
[Abstract]:Aim: to investigate the effects of bone marrow mesenchymal stem cell (BMSCs(Bone Marrow Mesenchymal Stem Cells) transplantation via rat tail vein on the recovery of nerve function and the expression of Nogo-A after acute spinal cord injury (SCI(Spinal cord injuryin) in rats. Methods: bone marrow mesenchymal stem cells were isolated, cultured and purified by whole bone marrow adherent method. The rat model of acute spinal cord injury was established by modified Allen's method. 96 adult female Sprague-Dawley rats were randomly divided into 4 groups (24 rats / group), which were treated with BMSCs in sci group, sci group and BMSCs group. Before transplantation of bone marrow mesenchymal stem cells (BMSCs), carboxyfluorescein Succinimidyl Succinimidyl (BMSCs) was labeled with carboxyfluorescein Succinimidyl (BMSCs). The mice were injected with BMSCs suspension 1ml(5 脳 106 BMSCs in caudal vein of mice. The neurobehavioral function was evaluated by BBB neurobehavioral score on the 1st, 3rd, 7th and 14th day after operation, and the recovery of neurobehavioral function was observed. The expression of Nogo-A in spinal cord was measured by immunohistochemistry and RT-PCR method. Results after the successful transplantation of BMSCs, the positive CFSE staining cells were observed in the peripheral tissue of spinal cord injury nerve. The evaluation of nerve function in the positive CFSE staining group was significantly better than that in the control group on the 7th and 14th day after injury, and that in the solvent control group was better than that in the control group. Compared with the control group and the solvent control group, the expression of Nogo-A in peripheral nerve tissue of BMSCs group was significantly decreased on the 3rd, 7th and 14th day after operation. Conclusion: transplantation of bone marrow mesenchymal stem cells in rats with acute spinal cord injury can promote the recovery of spinal cord nerve function after injury, which is partly related to the inhibition or down-regulation of Nogo- production in spinal cord tissue by bone marrow mesenchymal stem cells.
