miRNA-363通过调控SOX4影响骨肉瘤细胞生长及凋亡

【摘要】:目的:探讨骨肉瘤组织中miRNA-363及SOX4的表达水平,以及miRNA-363对骨肉瘤细胞活力、细胞周期以及凋亡的影响及机制。方法:用real-time PCR法检测63例骨肉瘤患者肿瘤组织与癌旁组织中miRNA-363及SOX4的表达水平及相关性。用脂质体法将miRNA-363 mimics瞬时转染入MG-63人骨肉瘤细胞,检测细胞中miRNA-363及SOX4的表达水平;CCK-8实验检测各组细胞的活力;流式细胞术检测各组细胞的周期及凋亡率;检测转染SOX4 siRNA或质粒后miRNA-363的表达水平。结果:骨肉瘤组织miRNA-363表达明显低于瘤旁组织,SOX4的mRNA表达明显高于瘤旁组织,miRNA-363与SOX4的表达水平呈明显负相关。转染miRNA-363 mimics后MG-63细胞miRNA-363的表达水平明显上调,SOX4 mRNA及蛋白的表达水平明显下降;同时,细胞活力明显下降细胞周期阻滞在G_0/G_1期,凋亡率增高。改变SOX4表达后,miRNA-363的表达无明显变化,升高SOX4表达能够恢复miRNA-363抑制的细胞活力。结论:miRNA-363在骨肉瘤中呈低表达水平,过表达miRNA-363可能通过调控SOX4抑制骨肉瘤细胞的生长,并促进细胞凋亡。
[Abstract]:Aim: to investigate the expression of miRNA-363 and SOX4 in osteosarcoma, and the effect of miRNA-363 on the viability, cell cycle and apoptosis of osteosarcoma cells. Methods: the expression of miRNA-363 and SOX4 in 63 cases of osteosarcoma were detected by real-time PCR method. MiRNA-363 mimics was transiently transfected into MG-63 human osteosarcoma cells by liposome method. The expression levels of miRNA-363 and SOX4 were detected by CCK-8 assay, and the cell cycle and apoptosis rate were detected by flow cytometry. The expression level of miRNA-363 was detected after transfection of SOX4 siRNA or plasmid. Results: the expression of miRNA-363 in osteosarcoma was significantly lower than that in the adjacent tissues of osteosarcoma. The expression of miRNA-363 and SOX4 was negatively correlated with the expression of SSOX4 in paraneoplastic tissues. After transfection of miRNA-363 mimics, the miRNA-363 expression level of MG-63 cells decreased significantly, and the cell viability decreased significantly, and the cell cycle arrest in G_0/G_1 phase, and the apoptosis rate increased. There was no significant change in the expression of miRNA-363 after the change of SOX4 expression, but the increased expression of SOX4 could restore the cell viability inhibited by miRNA-363. Conclusion the expression of miRNA-363 is low in osteosarcoma. Overexpression of miRNA-363 may inhibit the growth of osteosarcoma cells and promote apoptosis by regulating SOX4.
