CREPT在脂肪肉瘤中的表达及对SW872细胞增殖的影响

【摘要】：目的 1、分析CREPT在腹膜后脂肪肉瘤中表达及对预后的影响;2、分析CREPT对SW872细胞增殖能力的影响。方法 1、收集2007年6月至2014年3月我院收治的腹膜后脂肪肉瘤患者的临床资料和肿瘤蜡块,并对患者预后进行随访。通过病理蜡块连续切片HE染色和免疫组化染色,分析CREPT在脂肪肉瘤的表达及预后的影响。2、采用RNA干扰(RNAi)技术将CREPT进行表达抑制,通过MTT法、细胞平板克隆实验、划痕实验检测CREPT对SW872细胞增殖的影响。结果CREPT在脂肪肉瘤具有高表达,其表达强弱与肿瘤的级别(χ2=67.070,P0.01)、核分裂数(χ2=52.055,P0.01)、肿瘤坏死(χ2=25.546,P0.01)、肿瘤分化(χ2=57.109,P0.01)明显相关,Log Rank检验发现,CREPT表达强弱(χ2=37.759,P0.01),核分裂数(χ2=71.644,P0.01),肿瘤分化(χ2=30.357,P0.01),肿瘤坏死(χ2=27.369,P0.01),肿瘤级别(χ2=51.374,P0.01),手术方式(χ2=11.178,P0.01)是影响患者生存时间的主要因素。Cox多因素生存分析显示CREPT表达(RR=2.289,P=0.015)和核分裂(RR=2.444,P=0.005)数是预后的独立影响因素。通过MTT、平板克隆实验、划痕实验证明CREPT对SW872细胞增殖具有促进作用,抑制CREPT表达,细胞增殖能力减弱(P0.05)。结论CREPT在脂肪肉瘤中高度表达,且与预后明显相关。抑制CREPT表达，SW872细胞增殖能力明显减弱。
[Abstract]:Objective 1. To analyze the expression of CREPT in retroperitoneal liposarcoma and its influence on prognosis, and to analyze the effect of CREPT on the proliferation of SW872 cells. Methods 1. The clinical data and tumor wax of retroperitoneal liposarcoma treated in our hospital from June 2007 to March 2014 were collected and the prognosis was followed up. The expression and prognosis of CREPT in liposarcoma were analyzed by HE staining and immunohistochemical staining. The expression of CREPT was inhibited by RNA interference technique. The expression of CREPT was inhibited by MTT method. The effect of CREPT on proliferation of SW872 cells was detected by scratch test.
