小鼠脊髓损伤后对少突胶质前体细胞的影响

【摘要】:在中枢神经系统中,少突胶质细胞是一类包裹在神经元轴突形成髓鞘化的神经细胞,在脊髓损伤以后,大部分的少突胶质细胞细胞和神经元都坏死,接着星形胶质细胞大量增殖形成胶质疤痕。胶质疤痕会阻碍神经元的再修复,以及髓鞘的修复。我们想研究OPC在脊髓损伤后,如何参与脊髓损伤后的自我修复。并且在以往的研究中,通过移植少突胶质细胞细胞前体来治疗脊髓损伤时,发现大部分的少突胶质细胞前体在体内分化成为星形胶质细胞。我们推测在脊髓损伤以后,体内微环境发生了改变,可能产生了部分因子,影响少突胶质细胞前体分化成为星形胶质细胞。我们通过PDGFRa-creER/Rosa-td-tTomato小鼠为研究对象,PDGFRa为少突胶质细胞前体的标志蛋白,通过tomato标记后,研究脊髓损伤以后,对少突胶质细胞前体发育的影响。我们发现在脊髓损伤后,被Tomato信号标记的细胞,一部分表达cc1阳性,然后有一小部分细胞表达GFAP阳性。表明大部分OPC参与形成髓鞘的修复,但是有一小部分OPC分化成为星形胶质细胞形成胶质疤痕。
[Abstract]:In the central nervous system, oligodendrocytes are a group of nerve cells that form myelination around the axons of neurons. After spinal cord injury, most of the oligodendrocytes and neurons are necrotic. Astrocytes then proliferate in large numbers to form glial scars. Glial scars block the rerepair of neurons and the repair of myelin sheath. We want to study how OPC participates in self-repair after spinal cord injury. In previous studies, it was found that most of the oligodendrocyte precursors differentiated into astrocytes in vivo after transplantation of oligodendrocyte precursors to treat spinal cord injury. We speculate that the microenvironment changes after spinal cord injury, which may produce some factors and influence the differentiation of oligodendrocytes into astrocytes. In this study, PDGFRa-creER/Rosa-td-tTomato mouse was used as a marker protein of oligodendrocyte precursor. The effects of tomato labeling on the development of oligodendrocyte precursor were studied after spinal cord injury. We found that after spinal cord injury, some cells labeled with Tomato signal expressed cc1 positive, and then a small number of cells expressed GFAP positive. It was suggested that most of OPC were involved in the repair of myelin sheath, but a small number of OPC differentiated into astrocytes to form glial scars.
