沉默Aurora-B抑制骨肉瘤细胞143B在裸鼠体内生长

【摘要】:目的:初步探讨沉默Aurora-B基因对人骨肉瘤细胞143B在裸鼠体内生长的影响。方法:构建人骨肉瘤143B细胞裸鼠移植瘤模型18只,随机分为3组(n=6):分别为LV-sh Aurora-B(AURKB)注射组(实验组)、LV Negative注射组(阴性对照组)和PBS注射组(空白对照组)。每3天(间隔2天)于肿瘤组织局部注射(共注射3次)。每7天皮外测量瘤体大小,5周后处死,取出瘤体,测量瘤体大小及质量,TUNEL法检测细胞凋亡指数。结果:LV-sh Aurora-B注射组瘤体生长速度显著低于LV Negative注射组及PBS注射组(P0.05),LV Negative注射组与PBS注射组之间比较无显著统计学差异;5周后,LV-sh Aurora-B注射组瘤体大小、质量显著低于LV Negative注射组及PBS注射组,LV Negative注射组与PBS注射组之间比较无显著统计学差异。LV-sh Aurora-B注射组瘤体组织细胞凋亡指数显著高于LV Negative注射组及PBS注射组(P0.05),LV Negative注射组与PBS注射组之间比较无显著统计学差异。结论:在裸鼠体内,沉默Aurora-B基因能有效诱导人骨肉瘤143B细胞凋亡,抑制人骨肉瘤143B细胞增殖。
[Abstract]:Aim: to investigate the effect of silencing Aurora-B gene on the growth of human osteosarcoma cell line 143B in nude mice. Methods: eighteen nude mice transplanted with human osteosarcoma 143B cells were randomly divided into three groups: LV-sh Aurora-BU AURKB injection group (experimental group) (negative control group) and PBS injection group (PBS injection group). The tumor tissue was injected locally every 3 days (2 days interval) (3 times). The tumor size was measured every 7 days and killed after 5 weeks. The tumor size and mass were measured and the apoptosis index was detected by Tunel. Results there was no significant difference in tumor growth rate between LV-sh Aurora-B injection group and PBS injection group and between PBS injection group and PBS injection group. After 5 weeks, the tumor size in LV-sh Aurora-B injection group was significantly lower than that in LV-Aurora-B injection group. There was no significant difference between the LV-sh Aurora-B injection group and the PBS injection group. The apoptotic index of the tumor tissue in the LV-sh Aurora-B injection group was significantly higher than that in the LV Negative injection group and the PBS injection group, and the LV-sh Aurora-B injection group was significantly higher than that in the LV Negative injection group and the PBS injection group. There was no significant difference between the ejection group and the PBS injection group. Conclusion: in nude mice, silencing Aurora-B gene can effectively induce apoptosis of human osteosarcoma 143B cells and inhibit the proliferation of human osteosarcoma 143B cells.
