爆炸致家兔急性肺损伤及血必净的保护作用

【摘要】：目的探讨血必净对爆炸致家兔急性肺损伤(ALI)的保护作用及其机制。方法健康清洁级家兔32只,随机分成正常对照组、ALI组和低剂量血必净组、高剂量血必净组,每组8只。爆炸致伤家兔,建立ALI动物模型后,低剂量血必净组、高剂量血必净组分别注射10、20 ml/kg血必净溶液,正常对照组、ALI组注射生理盐水。4 h采集血液,24 h采集支气管肺泡灌洗液(BALF)和肺组织标本,Western blot法和反转录PCR法测定肺组织MMP-9蛋白和mRNA表达水平,ELISA法测定BALF及血清中肿瘤坏死因子(TNF)-α、白介素(IL)-6、IL-10的含量,并测定肺湿干重比(W/D),光镜下观察肺组织病理学变化。结果 ALI组MMP-9、TNF-α、IL-6、IL-10表达量以及W/D均较正常对照组升高(P0.05)。经血必净治疗后,低剂量血必净组、高剂量血必净组的MMP-9、TNF-α、IL-6表达量以及W/D均较ALI组降低(P0.05),IL-10表达量较ALI组升高(P0.05),均以高剂量组改变最显著(P0.05)。肺病理切片显示ALI组肺泡及间质水肿,肺泡腔内有浆液性渗出物及大量炎性细胞、红细胞渗出,低剂量血必净组、高剂量血必净组肺损伤程度明显减轻。结论血必净可通过降低MMP-9的表达抑制失控性炎症反应,减轻肺损伤,保护肺组织。
[Abstract]:Objective to investigate the protective effect and mechanism of Xuebijing on acute lung injury (Ali) induced by explosion in rabbits. Methods Thirty-two healthy clean grade rabbits were randomly divided into two groups: the normal control group, the control group and the low-dose Xuebijing group, and the high-dose Xuebijing group with 8 in each group. Rabbits were injured by explosion. After Ali animal model was established, low dose Xuebijing group and high dose Xuebijing group were injected with 10 ~ 20 ml/kg Xue Bi Jing solution, respectively. Determination of MMP-9 protein and mRNA expression in lung tissue by Western blot and reverse transcription-polymerase chain reaction (RT-PCR) in 24 h after injection of normal saline for 24 h and 24 h after injection of normal saline in control group. Elisa method for detection of MMP-9 protein and mRNA expression in serum and BALF in serum and bronchoalveolar lavage fluid (bronchoalveolar lavage fluid) and lung tissue samples were determined by reverse transcription-polymerase chain reaction (RT-PCR). Tumor necrosis factor (TNF)-伪, interleukin (IL)-6 and IL-10, Lung wet dry weight ratio (W / D) was measured and lung histopathology was observed under light microscope. Results in Ali group, the expression of MMP-9, TNF- 伪, IL-6, IL-10 and W- / D in Ali group were higher than those in normal control group (P0.05). After treatment with Jingxuebijing, the expression of MMP-9 TNF- 伪 -IL-6 and W- / D in low dose Xuebijing group and high dose Xuebijing group were lower than that in Ali group (P0.05), the expression of IL-10 was higher than that in Ali group (P0.05), and the most significant change was in high-dose group (P0.05). Lung pathological sections showed alveolar and interstitial edema in Ali group, serous exudates and inflammatory cells in alveolar cavity, exudation of red blood cells, reduction of lung injury in low dose Xuebijing group and high dose Xuebijing group. Conclusion Xuebijing can attenuate lung injury and protect lung tissue by reducing the expression of MMP-9.
