硫化氢对烧伤大鼠体外培养皮肤巨噬细胞MAPK信号传导通路中ERK、JNK及p38 mRNA的影响

【摘要】:目的研究H2S对烧伤大鼠体外培养皮肤巨噬细胞MAPK信号传导通路中ERK、JNK及p38 m RNA表达水平的影响,从而探究H2S参与创面愈合的分子机制。方法把烧伤大鼠皮肤巨噬细胞作为研究对象,从30只健康SD大鼠中选取5只大鼠不做任何处理,剩余25只大鼠制造深Ⅱ°烧伤模型(经病理切片证实)。从5只正常大鼠中随机选取1只作为空白对照组,从25只烧伤大鼠中选取12只随机分为4组:烧伤对照组、Na HS组、格列本脲(GLBN)组、Na HS+格列本脲组,每组3只。造模后分离正常或烧伤大鼠背部皮肤基底巨噬细胞至培养瓶。待稳定培养后,分别加入终浓度为200umol/L的相应药物给予干预(空白对照组和烧伤对照组加入等量的PBS溶液作为对照),在药物接触1h、6h、12h每个时相点利用RT-PCR技术检测巨噬细胞中ERK、p38、JNK的m RNA含量变化。结果1、各组ERK m RNA表达结果进行比较:(1)烧伤对照组与空白对照组在ERK m RNA转录水平上有统计学差异(P0.05);(2)在同一时相点,Na HS组与各组别在ERK m RNA转录水平上有统计学差异(P0.05);(3)在同一时相点,Na HS+GLBN组与GLBN组在ERK m RNA转录水平上统计学有差异(P0.05);(4)在同一时相点,GLBN组与烧伤组在ERK m RNA转录水平上比较有差异(P0.05)。2、各组JNK m RNA表达结果进行两两比较:(1)烧伤组与空白对照组在JNK m RNA转录水平上有统计学差异(P0.05);(2)在同一时间,Na HS组与各组别在JNK m RNA转录水平上有差别(P0.05);(3)在同一时相点,Na HS+GLBN组与GLBN组JNK m RNA表达量差别比较有统计学意义(P0.05);(4)在同一时相点,GLBN组与烧伤组在JNK m RNA转录水平上比较有差异(P0.05)。3、各组p38 m RNA表达结果进行比较:(1)烧伤对照组与空白对照组在p38m RNA转录水平上有统计学差别(P0.05);(2)在同一时间点,Na HS组与各组别在p38 m RNA转录水平上有差别(P0.05);(3)在同一时间点,Na HS+GLBN组与GLBN组p38 m RNA表达量差异比较有统计学意义(P0.05);(4)在同一时间点,GLBN组与烧伤对照组在p38 m RNA转录水平上差别明显(P0.05)。结论硫化氢通过调控K+通道,使ERK m RNA相对表达水平升高,JNK和p38m RNA表达水平下降,从而影响烧伤大鼠体外培养皮肤巨噬细胞MAPK信号通路的传导。
