人参皂苷Rb1治疗兔耳增生性瘢痕相关机制的探索

【摘要】:目的:通过兔耳创建增生性疤痕模型,疤痕内注射药物人参皂苷Rb1,观察疗效及探索相关机理。方法:随机取8只大耳型新西兰白兔,每耳6处直径1cm圆形伤口,创建增生性疤痕模型,待其上皮化后设空白组(N),地塞米松阳性对照组(D),生理盐水对照组(S),人参皂苷Rb1组(R)。其中地塞米松组及人参皂苷组均应用生理盐水配置浓度为1mg/ml,疤痕内注射药物,每次0.3ml,每三天1次,共三次。注射后第1、2、4、8周随机收集2只兔子标本(24例)行HE染色观察瘢痕增生指数(SEI)、行Masson染色观察胶原增生情况、免疫组化法检测增值细胞核抗原(PCNA)表达、逆转录聚合酶链反应检测转化生长因子β(TGF-β)及Ⅰ型胶原表达。各组数据除以空白组数据进行数据标准化。软件使用SPSS19.0,计量资料以“x±s”表示,使用方差分析,若数据不符合正态性及方差齐性,改用非参数检验。选α=0.05,当P≤0.05,数据有统计学意义。结果:用药后第1、2周三组间未见统计学差异,第4、8周,D及R组较S组瘢痕增生改善明显,并随时间的延长疗效增加,SHI降低(P0.05),胶原增生面积减少(P0.05),PCNA表达降低(P0.05),TGF-β1及Ⅰ型胶原m RNA表达降低(P0.05),但D组及R组间未见统计学差异。结论:局部注射人参皂苷Rb1能抑制活性细胞增殖,血管生成减少,抑制TGF-β1表达,胶原合成减少,有效改善兔耳疤痕的增生,为使用人参皂苷Rb1防治疤痕形成提供理论基础。
[Abstract]:Aim: to establish a hypertrophic scar model in rabbit ear and inject ginsenoside Rb1 into the scar to observe the curative effect and explore the related mechanism. Methods: eight New Zealand white rabbits with large ears were randomly selected, with 6 diameter 1cm round wounds in each ear, and then the hypertrophic scar model was created. After epithelization, a blank group of (N), dexamethasone positive control group, (D), normal saline control group, (S), ginsenoside Rb1 group (R). Was set up.
