关节表面软骨与游离体表面软骨FT-IR光谱成像的对比研究

【摘要】:研究目的及背景: 以关节表面软骨及关节内游离体表面软骨为研究对象,通过FT-IR光谱成像及相关分析方法,探索同处于一个关节腔微环境中两种表面软骨在分子水平的差异性,更深层次分析游离体表面软骨生物学行为发生变化的特点,从光谱学角度研究游离体的形成因素及与关节炎的关系,探寻关节软骨生长、分化规律并为临床治疗提供思路和理论依据。 资料与方法: 1.临床资料吉林大学中日联谊医院关节外科2013年1月至2014年1月资料较完整的行髋膝关节置换术的患者作为实验组样本来源,术中可提取到关节内游离体标本。对照组样本来源为同一关节腔内的经病理证实为正常的关节软骨。其中髋关节骨性关节炎1例,膝关节骨性关节炎9例;男性4例,女性6例。所有患者均进行髋膝部位正侧位的放射学检查,,用以明确诊断。所有样本均进行组织病理学检查,确定组间的差异性。 2.方法所用样本经成像前处理之后经FT-IR成像仪系统成像,收集各样本的吸收谱线,并对谱线进行进一步处理和计算。 3.分析利用统计学软件(SPSS17.0)对两组的二阶导数谱线波数位置进行统计学分析;选取所有样本的胶原及蛋白多糖波数及其对应的红外吸收值组成矩阵进行主成分分析。 结果: 1.两种软骨中胶原分子的N-H弯曲振动波数位置对照组为1514cm-1,实验组表现出明显蓝移,为1516cm-1。 2.两种软骨归属为酰胺II的N-H弯曲振动和C-N的拉伸震动波数对照组为1573cm-1,实验组表现出明显的蓝移,波数位置为1575cm-1。 3.通过间接衡量两种样本的软骨中蛋白多糖总成分含量,两样本无差异。 4.主成分分析法发现两者在主成分坐标系的散点图中分布有差异性。 结论: 1.游离体表面软骨的胶原成分较关节表面软骨的分子更加稳定,表明游离体表面软骨成分中起骨架支撑作用胶原更不容易被胶原酶所分解; 2.同处于一个微环境下的关节表面软骨和游离体表面软骨的PG总成分相同,表明PG作为填充物未出现质的变化; 3.在胶原和蛋白多糖的成分组成上两者是有区别的,游离体表面软骨在成分上表现出明显的缺陷性。
[Abstract]:Objective and background: the articular surface cartilage and the articular free body surface cartilage were studied by FT-IR spectral imaging and correlation analysis. To explore the differences between the two kinds of surface cartilage at molecular level in the same microenvironment of articular cavity, and to further analyze the characteristics of the biological behavior of free surface cartilage. The formation factors of free body and the relationship between free body and arthritis were studied from the point of view of spectroscopy, the growth and differentiation of articular cartilage were explored and the theoretical basis for clinical treatment was provided. Data and methods: 1. Clinical data from January 2013 to January 2014, the patients undergoing hip and knee arthroplasty in Jilin University Sino-Japanese Friendship Hospital from January 2013 to January 2014 were selected as experimental group samples, and intraarticular free body specimens could be extracted during the operation. The control group came from the same articular cavity, pathologically proved to be normal articular cartilage. One case of hip osteoarthritis, 9 cases of knee osteoarthritis, male 4 cases, female 6 cases. All patients underwent radiographic examination of the hip and knee. All samples were examined by histopathology to determine the difference between groups. 2. 2. Methods the absorption lines of the samples were collected by FT-IR imaging system after pre-processing, and the spectral lines were further processed and calculated. Statistical software (SPSS17.0) was used to analyze the position of the second derivative line wavenumber of the two groups, and the wave numbers of collagen and proteoglycan and their corresponding infrared absorption values were selected for principal component analysis. Results: 1. The N-H bending vibrational wavenumber position of collagen molecules in the two kinds of cartilage was 1514cm-1 in the control group and 1516cm-1.2 in the experimental group. The wave number of N-H bending vibration and C-N tensile vibration were 1573 cm ~ (-1) and 1575 cm ~ (-1), respectively, which belonged to amide II, and the position of wave number was 1575 cm ~ (-1) in the experimental group. The total content of proteoglycan in cartilage of two kinds of samples was measured indirectly, and there was no difference between the two samples. 4. Principal component Analysis (PCA) shows that there are differences between them in scattered plot of principal component coordinate system. Conclusion: 1. The collagen composition of free cartilage is more stable than that of articular cartilage, which indicates that collagen is less likely to be decomposed by collagenase. The total composition of PG in articular surface cartilage and free body surface cartilage was the same as that in a microenvironment, indicating that there was no qualitative change in PG as filler. 3. The composition of collagen and proteoglycan were different, and the free surface cartilage showed obvious defect in composition.
