髓核组织高温相关丝氨酸蛋白酶A1表达水平与椎间盘退变程度的相关性

【摘要】:目的分析髓核组织高温相关丝氨酸蛋白酶A1(HtrA1)的表达水平与椎间盘退变程度(Pfirrmann分级)的相关性。方法 36例需手术摘除髓核组织的患者,术前行磁共振成像检查,据矢状位T2加权相磁共振对拟手术节段予Pfirrmann分级;采用逆转录PCR及Western blot方法检测髓核组织中HtrA1的mRNA及蛋白表达水平,对髓核组织HtrA1的表达水平与Pfirrmann等级进行相关性分析。结果 36例椎间盘经磁共振成像检查,PfirrmannⅠ级3例、Ⅱ级10例、Ⅲ级11例、Ⅳ级7例、Ⅴ级5例;髓核组织HtrA1 mRNA及蛋白表达水平随Pfirrmann等级增高而增加,经Bonferroni检验,除Ⅳ、Ⅴ级间外(P0.05),余各级间两两比较差异均有统计学意义(P0.05),经Spearman秩相关分析:HtrA1 mRNA及蛋白表达水平与Pfirrmann分级间存在秩相关关系(P0.000 1)。结论髓核组织HtrA1 mRNA及蛋白表达水平随Pfirrmann等级增高而升高,提示HtrA1与椎间盘退变程度存在相关性。
