睾酮对膝关节骨关节炎去势雄兔的软骨修复作用

【摘要】:目的探讨睾酮对膝关节骨关节炎(KOA)睾丸切除(去势)雄兔的软骨修复作用。方法将18只雄兔采用改良Hulth法建立右侧KOA模型并去势,8周时随机处死6只,确认模型建立成功。将剩余12只兔随机分为观察组和对照组,每组6只。观察组每隔2周肌注十一酸睾酮6 mg/kg,对照组同时肌注等量生理盐水,共8周(4次)。分组处理第8、12、16周,采用电化学发光免疫分析法检测血清睾酮;分组处理第16周末处死,取出右股骨髁,肉眼观察标本大体情况,采用免疫组化法检测软骨组织胰岛素样生长因子1(IGF-1)表达,并进行关节软骨Mankin's评分。结果两组第8周血清睾酮水平比较差异无统计学意义(P0.05),观察组第12、16周血清睾酮水平均明显高于对照组(P均0.05)。对照组右膝关节明显肿大,关节软骨面变薄、粗糙,软骨表面色泽变暗,溃疡较深,边缘骨赘形成明显。观察组右膝关节肿大程度轻于对照组,软骨表面有光泽,可见小溃疡和不规则裂隙。观察组软骨组织IGF-1阳性表达率高于对照组,Mankin's评分低于对照组(P均0.05)。结论睾酮可促进KOA去势雄兔的软骨修复;其机制可能与促进IGF-1表达有关。
[Abstract]:Objective to investigate the effect of testosterone on cartilage repair of (KOA) orchiectomy (ovariectomized) male rabbits with knee osteoarthritis. Methods the right KOA model was established by modified Hulth method in 18 male rabbits and 6 rabbits were randomly killed at 8 weeks. The remaining 12 rabbits were randomly divided into observation group (n = 6) and control group (n = 6). Testosterone undecanoate 6 mg/kg, was injected intramuscularly every 2 weeks in the observation group and the control group was intramuscularly injected with the same amount of normal saline for 8 weeks (4 times). Serum testosterone was detected by electrochemiluminescence immunoassay (ECLIA) at week 812 ~ (12). The right femoral condyle was removed from the right femoral condyle at the end of the 16th week of treatment. The gross situation of the specimens was observed with naked eyes. The expression of insulin-like growth factor 1 (IGF-1) in cartilage was detected by immunohistochemical method, and the articular cartilage Mankin's score was evaluated. Results there was no significant difference in serum testosterone levels between the two groups at the 8th week (P0.05). The serum testosterone levels in the observation group were significantly higher than those in the control group at the 16th week (P 0.05). In the control group, the right knee joint was obviously enlarged, the articular cartilage was thinned, rough, the color of the cartilage was darkened, the ulcers were deeper and the marginal osteophytes were formed obviously. The right knee joint in the observation group was slightly enlarged than that in the control group, the cartilage surface was lustrous, small ulcers and irregular cracks could be seen. The positive expression rate of IGF-1 was higher and the Mankin's score was lower in the observation group than that in the control group (P < 0. 05). Conclusion testosterone can promote cartilage repair in KOA ovariectomized rabbits and its mechanism may be related to the promotion of IGF-1 expression.
