滑膜间充质干细胞与软骨细胞移植修复膝关节软骨缺损

【摘要】:背景:膝关节软骨损伤后修复较困难。有研究发现将关节软骨细胞和滑膜间充质干细胞混合培养接种于壳聚糖支架材料中,均可以形成软骨基质。目的:观察滑膜间充质干细胞与软骨细胞与壳聚糖/Ⅰ型胶原复合支架材料对膝关节软骨缺损的修复情况。方法:将大鼠膝关节滑膜间充质干细胞及软骨细胞按1∶2比例共培养于壳聚糖/Ⅰ型胶原复合支架材料中,将此复合材料移植于膝关节软骨缺损模型大鼠膝关节缺损处进行修复。结果与结论:(1)组织学变化:苏木精-伊红染色、甲苯胺蓝染色及番红O染色显示,植入后4,8,12周,大鼠膝关节软骨细胞逐渐增多;(2)软骨样组织标记物Ⅱ型胶原表达:免疫组织化学染色显示,移植后4,8,12周,大鼠膝关节软骨组织细胞及细胞基质可见Ⅱ型胶原阳性表达。(3)结果证实,滑膜间充质干细胞与软骨细胞混合培养于壳聚糖/Ⅰ型胶原复合支架材料中,在体内环境下能够形成软骨样组织,有利于损伤膝关节软骨缺损的修复。
[Abstract]:Background: repair of articular cartilage is difficult after injury. It was found that chondrocytes and synovial mesenchymal stem cells could be cultured in chitosan scaffold to form cartilage matrix. Aim: to observe the repair of cartilage defect of knee joint with synovial mesenchymal stem cells chondrocytes and chitosan / type I collagen composite scaffold. Methods: rat synovial mesenchymal stem cells and chondrocytes were co-cultured in chitosan / type I collagen composite scaffold at 1:2 ratio. The composite material was transplanted to the knee joint defect of the rat model of knee cartilage defect for repair. Results and conclusion: (1) histological changes: hematoxylin eosin staining toluidine blue staining and Phanerin O staining showed that the number of chondrocytes in the knee joint of rats gradually increased at 812 weeks after implantation.
