含稀土镧电纺纳米纤维膜修复家兔全层皮肤缺损

【摘要】:目的:探究聚己内酯-明胶-氯化镧(PCL-GE-LaCl_3)电纺纳米纤维膜,在修复家兔全层皮肤缺损模型的作用。方法:利用静电纺丝技术制备PCL-GE、PCL-GE-LaCl_3(0.4%组)、PCL-GE-LaCl_3(0.8%组)三种含镧离子Wt%不同的膜。将27只成年家兔(雌雄不限)用于创面造模,于每只背部两侧作3对两两对称的2.50.×2.5cm大小全层皮肤缺损创面6个,将每只家兔6个创面随机分为3组:凡士林对照组,PCL-GE膜组,PCL-GE-LaCl_3实验组。对照组覆盖普通凡士林纱布,PCL-GE组覆盖PCL-GE膜,实验组分别覆盖PCL-GE-LaCl_3(0.4%)膜、PCL-GE-LaCl_3(0.8%)膜。术后3天、7天、15天每个时间点,凡士林对照组,PCL-GE膜组,PCL-GE-LaCl_3(0.4%组)膜、PCL-GE-LaCl_3(0.8%组)的实际创面数分别为6、6、8、8,术后各组各时间点观察创面,计算创面闭合率,取局部组织做病理切片、HE染色,统计分析。结果:(1)PCL-GE、PCL-GE-LaCl_3(0.4%组)、PCL-GE-LaCl_3(0.8%组)三)种电纺纳米纤维膜,三种纤维膜表面虽存在少量颗粒状物,但纤维均较为连续、均一,且呈现出两种纤维交错分布的结构,有较多孔隙。(2)PCL-GE膜的分解峰对应的温度大于475℃,PCL-GE-LaCl_3(0.4%组)分解峰对应温度为475℃、PCL-GE-LaCl_3(0.8%组)分解峰对应温度为400℃,可见随着镧离子含量的增大,复合纳米纤维膜的分解起始温度下降,表明镧离子加入PCL-GE中降低了复合纳米纤维材料的热稳定性,易于分解。(3)术中观察到电纺膜均与创面贴连紧密,呈半透明状。术后3d,电纺膜仍与创面贴连紧密,表面干燥,凡士林组仍可见少量渗出。术后7d,含镧纳米纤维膜表面仍较洁净,膜下肉芽形成。术后15d创面基完成上皮化,表面无材料残留迹象。(4)术后3天,PCL-GE-LaCl_3(0.4%组)创面闭合合率显著高于凡士林对照组,PCL-GE-LaCl_3(0.8%组)闭合率明显高于凡士林对照组及PCL-GE组。术后7天和15天,与凡士林对照组相比,其他组无明显差异。结论:1、PCL-GE-LaCl_3纳米纤维膜具有三维立体形态,有较高的孔隙率。2、随着镧离子的增多,PCL-GE-LaCl_3电纺膜的热稳定性下降,易于分解。3、PCL-GE-LaCl_3纳米纤维膜具有良好的亲水性,生物组织相容性。4、含镧电纺膜能减少创面分泌物,可能具有局部抗炎作用。5、在创面闭合进程早期,PCL-GE-LaCl_3(0.8%组)及PCL-GE-LaCl_3(0.4%组)电纺膜可能具有促进创面收缩的作用,以PCL-GE-LaCl_3(0.4%)组最明显。
[Abstract]:Objective: to investigate the effect of polycaprolactone gelatin lanthanum chloride (PCL-GE-LaCl_3) electrospun nanofiber membrane on repairing full-thickness skin defect in rabbits. Methods: three kinds of Wt% membranes containing lanthanum ions, PCL-GE,PCL-GE-LaCl_3 (0.4% group) and PCL-GE-LaCl_3 (0.8% group), were prepared by electrospinning. 27 adult rabbits (male and female) were used to establish wound models. Three pairs of pairwise 2.50. 脳 2.5cm full-thickness skin defect wounds were made on both sides of each back. Six wounds in each rabbit were randomly divided into three groups: Vaseline control group, PCL-GE membrane group and PCL-GE-LaCl_3 experimental group. The control group was covered with ordinary Vaseline gauze, the PCL-GE group was covered with PCL-GE membrane, and the experimental group was covered with PCL-GE-LaCl_3 (0.4%) membrane and PCL-GE-LaCl_3 (0.8%) membrane, respectively. At each time point of 3 days, 7 days and 15 days after operation, the actual number of wounds in Vaslin control group, PCL-GE membrane group, PCL-GE-LaCl_3 (0.4% group) membrane and PCL-GE-LaCl_3 (0.8% group) was 6, 6, 8, 8. the wound closure rate was calculated, local tissue was taken for pathological section, HE staining and statistical analysis. Results: (1) PCL-GE,PCL-GE-LaCl_3 (0.4% group), PCL-GE-LaCl_3 (0.8% group) electrospun nanofiber membranes, although there were a small amount of particles on the surface of the three kinds of fiber membranes, the fibers were continuous and uniform, and showed two kinds of fibers interlaced structure, with more pores. (2) the decomposition peak of PCL-GE film corresponding to the temperature is more than 475C, The corresponding temperature of decomposition peak of PCL-GE-LaCl_3 (0.4% group) is 475 鈩,
