U0126抑制铁死亡修复脊髓损伤的实验研究

【目的】脊髓损伤(Spinal Cord Injury,SCI)是脊柱损伤的严重并发症,具有高发病率、高致残率、高致死率以及并发症多等特点,损伤后神经功能的恢复是亟待解决的世界性难题。脊髓损伤后局部抑制性微环境的形成导致大量神经元及各种支持细胞的死亡,加重继发性损伤。本研究团队前期研究表明,脊髓继发性损伤阶段,存在一种新定义的程序性细胞死亡模式-铁死亡(Ferroptosis),其不同于传统意义上的坏死、凋亡、坏死性凋亡和焦亡,能够促进神经元等细胞的死亡,参与继发性损伤。本课题旨在明确铁死亡参与脊髓继发性损伤病理机制,并通过铁死亡抑制剂U0126干预,探讨其对脊髓损伤修复的有效性,并进一步明确脊髓损伤的病理机制及新的修复靶点。【方法】1.脊髓继发性损伤阶段铁死亡标志物检测将雌性Wistar大鼠(240 g±10 g,10周龄)随机分为假手术组、损伤组。假手术组大鼠单纯摘除T10节段椎板,损伤组大鼠采用NYU IMPACTOR MODEL-Ⅱ脊髓打击器制作Wistar大鼠脊髓损伤挫伤模型。分别于手术后6 h采用Western Blotting、GSH检测试剂盒等方法检测手术后各组铁死亡标...
