丙泊酚通过上调miR-219-5p抑制肝癌生长和转移的作用与机制研究

研究背景原发性癌是常见的恶性肿瘤,手术切除是目前治疗肝癌最常用和最有效的方法,然而即使是根治性手术切除的患者,5年的复发率仍高达70%。围术期是癌细胞转移的关键时间窗,丙泊酚作为肿瘤切除手术常用的静脉麻醉药,研究显示其具有抑制肿瘤生长和转移的作用。丙泊酚抑制肝癌的具体作用及机制目前国内外尚无明确报道。mniRNAs在肿瘤发生发展过程中发挥着重要作用,通过前期实验发现,丙泊酚可显著上调miR-219-5p表达,miR-219-5p在多种肿瘤中均具有抑癌作用。因此我们探究丙泊酚是否通过上调miR-219-5p的表达抑制肝癌的生长与转移,并就具体作用机制作进一步探讨。研究目标1.验证丙泊酚通过上调miR-219-5p的表达抑制肝癌细胞增殖与侵袭转移。2.探讨丙泊酚上调miR-219-5p抑制肝癌侵袭转移与EMT进程的作用机制,初步探讨miR-219-5p的作用靶基因RAB18。3.揭示RAB18在人肝癌细胞系、肝癌组织及其匹配癌旁肝组织中的表达水平,及其在肝癌患者预后评估的价值;探讨肝癌发生发展过程中RAB18的功能及调控机制,以寻求肝癌侵袭转移新的理论依据。研究内容与方法1.揭示丙泊酚通过...
