青蒿素抑制大鼠角膜移植排斥反应

【摘要】:目的探讨全身给予青蒿素,研究其对大鼠角膜移植排斥反应的抑制作用。方法本实验通过把Wistar大鼠和SD大鼠作为移植给予和接受的对象,建立角膜移植的动物模型。随机分为4组,正常组:不行角膜手术,也不药物干预;阳性对照组:给予1%羧甲基纤维素钠(CMC-Na)溶液1.5 m L;环孢霉素A组:给予环孢霉素A 10 mg/kg;青蒿素组:给予青蒿素600 mg/kg。术后在显微镜下观察各组角膜植片的状态,记录生存时间及排除反应指数,HE染色观察角膜植片的病理变化。结果阳性对照组、环孢霉素A组、青蒿素组角膜植片的平均存活时间分别为(11.67±0.67)d、(20.50±0.76)d、(18.67±0.67)d,环孢霉素A组、青蒿素组与阳性对照组比较,角膜植片的存活时间都明显增加。病理学检查结果:对照组可见角膜水肿,厚度增加,有较多的炎症细胞,周围可见新生血管,环孢霉素A组、青蒿素组的角膜植片均较阳性对照组明显好转,水肿程度减轻、有少量炎性细胞浸润,效果明显好于阳性对照组。结论全身给予青蒿素,可以使移植术后角膜植片存活的时间延长,使角膜的水减轻肿,减少炎细胞的浸润,从而能够抑制移植免疫排斥反应的发生。
[Abstract]:Objective to investigate the inhibitory effect of artemisinin on corneal allograft rejection in rats. Methods Wistar rats and SD rats were used as recipients. Animal models of corneal transplantation were established. They were randomly divided into 4 groups: normal group: no corneal surgery, no drug intervention; The positive control group was given 1% CMC-Na solution, the cyclosporine A group was given cyclosporine A 10 mg / kg, and the artemisinin group was given artemisinin 600 mg / kg. Results the mean survival time of corneal grafts in the positive control group, cyclosporine A group and artemisinin group were 11.67 卤0.67 d (20.50 卤0.76) and 18.67 卤0.67 days respectively, and those in cyclosporine A group were 18.67 卤0.67 days. The survival time of corneal grafts in the artemisinin group was significantly longer than that in the positive control group. Pathological examination showed that corneal edema was observed in the control group, the thickness increased, there were more inflammatory cells, neovascularization was seen around, and cyclosporine A group. The corneal grafts in the artemisinin group were better than those in the positive control group, the degree of edema was reduced, and a small amount of inflammatory cells infiltrated, the effect was obviously better than that in the positive control group. It can prolong the survival time of corneal grafts after transplantation, reduce the swelling of cornea, reduce the infiltration of inflammatory cells, and inhibit the occurrence of allograft rejection.
