小鼠急性中耳炎模型中中耳与内耳差异基因的筛选及基因网络分析

【摘要】:背景中耳炎是儿童时期最常见的感染性疾病之一,多种基因均参与其发病过程。目前已有关于小鼠中耳炎的基因芯片研究,但都是基于流感嗜血杆菌所致的中耳炎,对肺炎链球菌引起的中耳炎的基因表达谱芯片研究至今尚未开展过。并且目前对中耳炎研究主要倾向于传统单基因方面,显然已不能满足当前系统生物学研究的需要。因此,本研究一方面绘制出流感嗜血杆菌所致中耳炎的差异基因互作网络图,筛选出其主要网络中心节点,另一方面研究肺炎链球菌性小鼠急性中耳炎时所涉及的基因改变。方法用生物信息学软件对GEO数据库中已有流感嗜血杆菌中耳炎相关基因芯片原始数据进行分析,绘制差异基因网络;用鼓膜穿刺法建立小鼠肺炎链球菌中耳炎模型,并进行m RNA表达谱生物芯片分析,筛选中耳与内耳的差异基因。结果流感嗜血杆菌中耳炎表达谱芯片(GSE66141)经STRING在线工具、Cytoscape软件分析后得到其基因网络图;肺炎链球菌中耳炎表达谱芯片结果显示中耳中702个基因上调,250个基因下调;内耳中上调基因140个,下调基因171个。结论本研究的结果可能为中耳炎的分子机制提供更好的了解。
[Abstract]:Background Otitis media is one of the most common infectious diseases in childhood.Up to now, there has been gene chip research on mouse otitis media, but it is based on Haemophilus influenzae. The gene expression microarray of Streptococcus pneumoniae induced otitis media has not been carried out yet.At present, the research on otitis media is mainly focused on the traditional single gene, which obviously can not meet the needs of the current system biology research.Therefore, on the one hand, the differential gene interaction network of Haemophilus influenzae induced otitis media was drawn, and its main network center nodes were screened. On the other hand, the genetic changes involved in acute otitis media induced by Streptococcus pneumoniae in mice were studied.Methods the original data of Haemophilus influenzae otitis media related gene chip in GEO database were analyzed with bioinformatics software, and the differential gene network was drawn, and the mouse model of Streptococcus pneumoniae otitis media was established by tympanic membrane puncture.The differential genes of middle ear and inner ear were screened by biochip analysis of m RNA expression profile.Results the gene network of Haemophilus influenzae tympanitis expression microarray (GSE66141) was analyzed by STRING online tool Cytoscape software, and the results showed that 702 genes were up-regulated and 250 genes down-regulated in the middle ear of Streptococcus pneumoniae.There were 140 up-regulated genes and 171 down-regulated genes in the inner ear.Conclusion the results of this study may provide a better understanding of the molecular mechanism of otitis media.
