HMGB1和MMP-2、MMP-9在喉癌组织中的表达及其与预后的关系

【摘要】:目的探讨喉癌组织中高迁移率族蛋白B1(HMGB1)和基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)的表达及其与临床病理特征和预后的关系。 方法应用免疫组织化学EnVision二步法检测61例喉癌组织中HMGB1和MMP-2、MMP-9蛋白表达;实时荧光定量PCR法检测30例喉癌组织中HMGB1和MMP-2、MMP-9mRNA表达。 结果HMGB1和MMP-2、MMP-9蛋白在喉癌组织中的阳性表达率明显高于癌旁正常组织(χ2值分别44.934和49.923、36.054,P值均0.01);HMGB1和MMP-2、MMP-9mRNA在喉癌组织中的相对表达水平明显高于癌旁正常组织(t值分别为5.940和7.005、7.664,P值均0.01)。HMGB1和MMP-2、MMP-9蛋白表达均与喉癌T分期、临床分期和淋巴结转移有关(P值均0.05);HMGB1和MMP-9蛋白在喉癌中的表达呈正相关(r=0.381,P0.01),HMGB1和MMP-2蛋白在喉癌组织中的表达无相关性(r=0.094,P0.05);Kaplan-Meier生存分析结果显示,HMGB1和MMP-9在表达阳性患者的生存率低于表达阴性患者(χ2值分别为4.974、6.418,P值均0.05);COX回归分析显示,HMGB1是危险因子,其表达越高,患者预后越差(P0.05)。 结论HMGB1和MMP-2、MMP-9与喉癌的浸润、转移有关;HMGB1和MMP-9的表达有协同促进作用;HMGB1是影响预后的独立危险因素。
[Abstract]:Objective to investigate the expression of high mobility group protein B1 (HMGB1), matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in laryngeal carcinoma and its relationship with clinicopathological features and prognosis. Methods the expression of HMGB1 and MMP-2,MMP-9 in 61 cases of laryngeal carcinoma was detected by immunohistochemical EnVision two-step method, and the expression of HMGB1 and MMP-2,MMP-9mRNA in 30 cases of laryngeal carcinoma was detected by real-time fluorescence quantitative PCR. Results the positive expression rate of HMGB1 and MMP-2,MMP-9 protein in laryngeal carcinoma was significantly higher than that in adjacent normal tissues (蠂 ~ 2 = 44.934 and P = 49.923 ~ 36.054, respectively). The relative expression levels of HMGB1 and MMP-2,MMP-9mRNA in laryngeal carcinoma tissues were significantly higher than those in adjacent normal tissues (t = 5.940 and 7.005 ~ 7.664g, respectively, P < 0.01). The expression of HMGB1 and MMP-2,MMP-9 protein was related to T stage, clinical stage and lymph node metastasis of laryngeal carcinoma (P < 0. 05). There was a positive correlation between the expression of HMGB1 and MMP-9 in laryngeal carcinoma (r = 0.381p 0.01), but no correlation between the expression of HMGB1 and MMP-2 protein in laryngeal carcinoma (P 0.05), Kaplan-Meier survival analysis showed that the survival rate of HMGB1 and MMP-9 in positive expression was lower than that in negative expression (蠂 2 = 4.9746.41818 P respectively). COX regression analysis showed that HMGB1 was a risk factor, the higher its expression, the worse the prognosis (P0.05). Conclusion HMGB1 and MMP-2,MMP-9 are associated with invasion and metastasis of laryngeal carcinoma, HMGB1 and MMP-9 expression are synergistic and HMGB1 is an independent risk factor for prognosis.
