干扰血管生成素表达对糖尿病大鼠视网膜视细胞超微结构的影响

【摘要】:目的探讨慢病毒shRNA-血管生成素(ANG)载体干扰ANG表达对糖尿病大鼠视网膜视细胞超微结构的影响。方法将40只大鼠随机分为对照组、模型组、空载体组及shRNA-ANG组各10只,后三组给予一次性腹腔注射链脲佐菌素(STZ)建立糖尿病大鼠模型。造模成功后2周,空载体组及shRNA-ANG组分别向玻璃体腔注射空载体及shRNA-ANG慢病毒载体,其他两组注射等剂量生理盐水。造模成功14周时,摘除眼球分离视网膜,检测视网膜组织ANG mRNA和蛋白相对表达量,透射电镜观察各组视网膜视细胞超微结构。结果模型组、空载体组视网膜组织ANG mRNA相对表达量均高于对照组、shRNA-ANG组(P均0.01),shRNA-ANG组视网膜组织ANG蛋白相对表达量低于其他三组(P均0.01)。shRNA-ANG组视网膜神经细胞核、线粒体和膜盘超微结构的病变程度均较模型组、空载体组减轻。结论抑制糖尿病大鼠视网膜ANG表达可减轻视网膜视细胞超微结构的病变程度。
[Abstract]:Objective to investigate the effect of (ANG) vector interference of lentivirus shRNA- angiopoietin (shRNA-) on the ultrastructure of retinal visual cells in diabetic rats. Methods Forty rats were randomly divided into control group, model group, empty carrier group and shRNA-ANG group. The latter three groups were given intraperitoneal injection of streptozotocin (STZ) to establish diabetic rat model. Two weeks after the establishment of the model, empty vector and shRNA-ANG lentivirus vector were injected into the vitreous body cavity in the empty vector group and shRNA-ANG group, and the other two groups were injected with the same dose of normal saline. At 14 weeks after the model was successfully made, the eyes were removed to separate the retina, the relative expression of ANG mRNA and protein in the retina tissue was detected, and the ultrastructure of retinal visual cells in each group was observed by transmission electron microscope. Results the relative expression of ANG mRNA in model group and empty carrier group was higher than that in control group and shRNA-ANG group (P0.01). The relative expression of ANG protein in retinal tissue of shRNA-ANG group was lower than that of other three groups (P0.01). The pathological degree of retinal nerve cell nucleus, mitochondria and membrane disc ultrastructure in shRNA-ANG group was less than that in model group and empty carrier group. Conclusion inhibiting the expression of ANG in retina of diabetic rats can reduce the pathological degree of retinal ultrastructure.
