TNF-a、IL-6在实验性急性末端回肠炎中的表达

【摘要】:目的:探讨TNF-α、IL-6的表达水平在实验性急性末端回肠炎中的变化。 方法:选取30只健康成年的雄性杂种犬,运用随机数字抽样法分成6组:正常对照组(A组)、高温高湿高血糖组(B组)、正常血糖颅脑火器伤组(C组)、高血糖颅脑火器伤组(D组)、正常血糖湿热下颅脑火器伤组(E组)、高血糖湿热下颅脑火器伤组(F组),每组5只。将后四组(C、D、E、F组)给予手枪弹颅脑火器伤造模。在实验组(B、C、D、E、F组)共五组造模后,将犬立刻仰卧位固定在手术台上,并行气管插管、心电监护、心肺复苏,观测各组犬生命体征的变化。观察过程中,除心脏挤压及使用球囊呼吸器抢救外,不做其他处理。在无菌条件下,立即开腹,收集距回盲部1-3cm处的末端回肠组织。肉眼大体观察,石蜡包埋后行苏木精-伊红染色法(HE染色)制作病理切片,并在光学显微镜下观察其变化,采用逆转录-聚合酶链反应(RT-PCR)检测造模后0.5h、1h、3h的TNF-αmRNA、IL-6mRNA表达水平的变化。 结果: 1、各组实验犬肛温随时间的变化实验组(B、C、D、E、F组)犬的肛温在各时间点(10min、1h、2h、3h)均有升高,随着时间的推移,肛温逐渐升高,以F组对犬的肛温影响最大,实验组分别与正常组比较,差异有统计学意义(PO.05)。通过实验,湿热、高血糖、颅脑火器伤三重因素实验条件下的肛温变化较其他两两组合的双重实验条件更为剧烈,两两组合的双重实验条件较单纯因素的实验条件下肛温的变化剧烈。 2、末端回肠组织肉眼及镜下炎性评分肉眼观,实验组(B、C、D、E、F组)均以急性炎性反应为表现,且F组较其它组炎性反应严重;病理切片镜下示,实验组炎性细胞浸润增多,淋巴管、乳糜管扩张,绒毛变粗变短,表现为急性炎性反应,实验组分别与正常组比较,差异有统计学意义(PO.05)。各实验组出现炎性评分随时间的延长,其炎性损伤的程度逐渐加重。 3、末端回肠组织TNF-αmRNA的表达水平各实验组(B、C、D、E、F组)伤后0.5h、1h、3h,TNF-αmRNA在末端回肠组织的表达逐渐升高,以F组升高最大,实验组分别与正常组比较,差异有统计学意义(PO.05)。通过实验表明,TNF-α可能参与了实验性急性末端回肠炎的发病过程,与时间呈依赖性,随着末端回肠组织炎性损伤的加重,其表达量增加,具有明显促炎作用。 4、末端回肠组织IL-6mRNA的表达水平伤后0.5h、1h、3h,实验组(B、C、D、E、F组)IL-6mRNA在末端回肠组织的表达依次升高,且随着时间延长,升高数值越大,以F组最明显,实验组分别与正常组比较,,差异有统计学意义(PO.05)。通过实验说明,IL-6在实验性急性末端回肠炎的发病过程中与时间呈依赖性,但其整体表达水平均较TNF-αmRNA的少,IL-6可能在实验性急性末端回肠炎的发病过程中起着辅助促炎效应。 结论: 1、在实验性ATI中,随着时间的延长,末端回肠组织炎性损伤逐渐加重; 2、TNF-α、IL-6在实验性ATI中的表达呈时间依赖性; 3、TNF-α、IL-6协同参与了实验性ATI的发病过程。
[Abstract]:Objective : To investigate the expression level of TNF - 伪 and IL - 6 in experimental acute terminal ileitis .

Methods : Thirty healthy adult male hybrid dogs were randomly divided into six groups : normal control group ( group A ) , high - temperature high - humidity hyperglycemia group ( group B ) , normal blood sugar craniocerebral gunshot wound group ( group C ) , high blood glucose and craniocerebral gunshot wound group ( group D ) , normal blood glucose wet - heat and craniocerebral gunshot wound group ( group F ) .

Results :

1 . The anal temperature of dogs in experimental group ( B , C , D , E , F ) increased with time ( 10min , 1h , 2h , 3h ) .

2 . At the end ileum , the visual and microscopic inflammatory scores were observed . The experimental groups ( group B , C , D , E , F ) were characterized by acute inflammatory reaction , and the F group had more inflammatory response than other groups .
Under the pathological section , the inflammatory cell infiltration in experimental group increased , the lymphatic vessels , the breast erosions were dilated and the villi became shorter and shorter . The experimental group showed acute inflammatory reaction . The experimental group had significant difference compared with the normal group ( PO.05 ) . The inflammatory score of each experimental group increased with time , and the degree of inflammatory injury was gradually increased .

3 . The expression level of TNF - 伪 mRNA in the terminal ileum was 0.5 h , 1 h , 3 h , and TNF - 伪 mRNA increased gradually in the terminal ileum . The results showed that TNF - 伪 might be involved in the pathogenesis of experimental acute terminal ileitis .

4 . IL - 6 mRNA expression in terminal ileum was 0.5h , 1h , 3h . The expression of IL - 6 mRNA in the terminal ileum was increased in the experimental group . The results showed that IL - 6 had a statistically significant difference with time in the pathogenesis of experimental acute terminal ileitis , but the expression level of IL - 6 was lower than that of normal group . IL - 6 could play an auxiliary pro - inflammatory effect in the pathogenesis of experimental acute terminal ileitis .

Conclusion :

1 . In the experimental ATI , with the extension of time , the inflammatory injury of terminal ileum was gradually increased ;


2 . The expression of TNF - 伪 and IL - 6 in experimental ATI was time - dependent ;
