肝星状细胞自噬水平与细胞增殖、凋亡水平的相关性研究

【摘要】:目的:探讨3-甲基腺嘌呤(3-methyladenine,3-MA)抑制大鼠肝星状细胞(HSC-T6)自噬后对HSC-T6细胞活化、增殖、凋亡、周期的影响及可能的作用机制。方法:体外培养大鼠肝星状细胞HSC-T6,用不同浓度的3-MA处理,分为四个实验组,即空白对照组(0.0mmol/l),低浓度组(2.5mmol/l),中浓度组(5.0mmol/l),高浓度组(10.0mmol/l)。用MTT法观察细胞增殖活力;用流式细胞技术检测4组细胞的周期和凋亡情况;用蛋白印迹法(Western blot)分析不同浓度3-MA作用下各组HSC-T6细胞活化标志α-平滑肌肌动蛋白(α-SMA)、自噬标志蛋白LC3Ⅱ、细胞周期蛋白Cyclin A、CyclinB1以及凋亡关键调节蛋白Bcl-2、BAX的表达变化。结果:不同浓度的3-MA作用于HSC-T6细胞24h后,3-MA实验组的HSC-T6细胞增殖明显受到抑制,细胞凋亡率逐渐增高(均P0.05);S期及G2/M期细胞比例均明显增加(均P0.05);Western blot显示加入3-MA处理后HCS-T6细胞LC3Ⅱ、α-SMA、Cyclin A、CyclinB1、Bcl-2表达量均逐渐下降(均P0.05),BAX表达量逐渐上调(P0.05)。结论:(1)自噬抑制剂3-MA下调HSC-T6细胞的自噬水平,抑制LC3Ⅰ向LC3Ⅱ转化,同时抑制HSCs的增殖和活化效应;(2)3-MA抑制HSC-T6细胞自噬后可分别通过下调Cyclin A、Cyclin B1蛋白水平诱导HSCs细胞周期阻滞在S期、G2/M期;(3)3-MA抑制HSC-T6细胞自噬后可通过上调促凋亡蛋白BAX表达和下调抗凋亡蛋白Bcl-2的表达,升高BAX/Bcl-2的比率而诱导HSC-T6细胞凋亡。
