MDSCs与CXCR4/CXCL12轴在非酒精性脂肪性肝炎小鼠中的作用及其机制研究

第一部分构建CDAA饮食诱导的小鼠NASH模型目的:构建CDAA饮食诱导的NASH小鼠模型,为进一步的深入研究做基础。方法:选用6-8周龄雄性SPF级C57BL小鼠,经适应性喂养1周后分为正常普通饮食NF组、CDAA模型组、CSAA对照组直到第12周末结束。造模结束时,留取血清检测生化指标:丙氨酸氨基转化酶(ALT)、门冬氨酸氨基转移酶(AST)、甘油三酯(TG)、尿素氮(BUN)、血糖(Blood Glucose)、总胆固醇(T-CHO)、总胆红素(TBIL)的水平;肝组织经石蜡包埋HE染色,镜下观察小鼠肝组织病理变化,天狼猩红染色评价NASH模型纤维化程度。并应用NAFLD活动度积分(NAFLD activity score,NAS)评分判断NASH模型程度;ELISA方法检测正常组、对照组、模型组外周血炎性因子PGE2、IL-6、INF-γ水平。结果:(1)经纯化CDAA模型饮食、CSAA对照饮食组喂养小鼠12周末其平均体重,肝指数与正常组无显著差异(P>0.05)。模型组小鼠肝脏色泽变黄,边缘较钝,质地较硬,弹性差。模型组外周血清ALT、AST、Glucose较正常组均明...
