TGF-β1在BMSC移植可能对大鼠肝纤维及肝癌进程影响中的作用

　　目的建立骨髓间充质干细胞体外培养及扩增体系的基础上,探讨不同时期移植BMSC、移植时间长短以及移植途径不同对大鼠肝纤维化进程的影响:从TGF-β1的角度出发,探讨BMSC移植在肝纤维化进程中的作用机制以及可能在肝硬化后肝癌发生中的影响。方法1、应用密度梯度离心结合贴壁培养法分离纯化雄性SD大鼠BMSC,分别向成骨细胞,成脂肪细胞诱导分化进行鉴定。2、在四氯化碳诱导大鼠肝纤维化模型的第六周、第八周,将来自雄性SD大鼠的BMSC以5×10⁶/只的剂量经尾静脉注入同种雌性大鼠体内。分别在BMSC移植4周及6周后处死各组大鼠,评价干预组与对照组的生存状态,并用Scheuer炎症活动度评分法和Schmid M纤维化评分法评价肝脏病理情况。3、在四氯化碳诱导大鼠肝纤维化模型的第六周,将来自雄性SD大鼠的BMSC以5×10⁶/只的剂量经腹腔、经尾静脉及肝内注射三种途径移植入同种雌性大鼠体内。BMSC移植6周后分别处死各组大鼠,评价各组的生存状态,异形细胞发生率,Scheuer炎症活动度评分法和Schmid M纤维化评分法评价肝脏病理情况,免疫组化法检测α-...
