阿魏酸钠经RhoA/Rho-kinase信号通路对肝硬化大鼠门脉压力的作用及其机制

　　【背景及目的】门脉高压是使肝硬化病人死亡风险增高的重要因素之一。实验性肝硬化大鼠肝脏中RhoA及Rho-kinase表达水平明显高于正常大鼠，RhoA/Rho-kinase信号通道的激活导致血管平滑肌收缩并抑制其舒张，从而促进了门脉高压的发生。相关研究报道，阿魏酸能够降低胆固醇合成，后者合成过程的中间产物GGPP是RhoA活化的关键物质。本实验旨在研究阿魏酸对肝硬化大鼠门脉压力的作用，并探讨相关分子机制。【方法】用胆管结扎方法制作肝硬化大鼠模型。实验动物分为SHAM+NS、BDL+NS及BDL+SF三组，比较分析各组大鼠大体特征及血清肝功能指标。阿魏酸治疗对肝脏纤维化程度的影响通过显微镜下观察肝脏HE染色切片、透射电镜观察肝脏超微结构改变，以及免疫组化检测肝脏a-SMA表达等综合评定。免疫组化方法检测各组大鼠肝脏中RhoA、Rho-kinase及eNOS的表达。分离、培养原代大鼠HSC，另购入人肝星状细胞系LX-2，分别给予阿魏酸/阿魏酸+GGPP处理，流式细胞术检测各处理因素作用下细胞凋亡率。原位肝灌注方法测定大鼠原位肝灌注压力/肝内血管阻力，以及由低到高浓度梯度的甲氧明对各组大鼠肝...
