【摘要】：目的探讨髓源性抑制细胞(MDSCs)在鼻咽癌患者外周血中的表达水平及其临床意义,并在荷瘤小鼠体内探讨MDSCs可能的作用。方法采用流式细胞术分析49例鼻咽癌患者外周血及荷瘤小鼠脾脏MDSCs的比例,采用ELISA分析鼻咽癌患者及荷瘤小鼠中MDSCs对T细胞分泌细胞因子的影响。结果鼻咽癌患者外周血中MDSCs的比例显著高于健康对照组,并且鼻咽癌患者外周血中MDSCs的比例与肿瘤的分期有关,晚期患者外周血中MDSCs的比例高于早期患者;鼻咽癌荷瘤小鼠脾脏内MDSCs的数量明显高于对照组;研究表明MDSCs可明显抑制T细胞分泌细胞因子。结论鼻咽癌患者外周血中MDSCs的比例较健康人高,且与鼻咽癌分期密切相关,荷瘤小鼠实验表明肿瘤组小鼠脾脏MDSCs的数量明显高于对照组,并且MDSCs可影响荷瘤小鼠细胞因子的分泌。MDSCs可能作为鼻咽癌生物治疗的1个靶点。
[Abstract]:Objective to investigate the expression of myelogenous inhibitory cell (MDSCs) in peripheral blood of patients with nasopharyngeal carcinoma (NPC) and its clinical significance. Methods flow cytometry was used to analyze the proportion of MDSCs in peripheral blood and spleen of 49 patients with nasopharyngeal carcinoma. ELISA was used to analyze the effect of MDSCs on T cell cytokines secretion in nasopharyngeal carcinoma patients and tumor-bearing mice. Results the percentage of MDSCs in peripheral blood of NPC patients was significantly higher than that of healthy controls. The proportion of MDSCs in peripheral blood of patients with nasopharyngeal carcinoma was related to the stage of tumor, and the proportion of MDSCs in peripheral blood of patients with advanced stage was higher than that of patients with early stage. The amount of MDSCs in the spleen of nasopharyngeal carcinoma bearing mice was significantly higher than that of control group. Conclusion the proportion of MDSCs in peripheral blood of patients with nasopharyngeal carcinoma is higher than that of healthy people, and it is closely related to the stage of nasopharyngeal carcinoma. Tumor-bearing mice experiment showed that the number of spleen MDSCs in the tumor group was significantly higher than that in the control group. And MDSCs can affect cytokine secretion in tumor-bearing mice. MDSCs may be a target for nasopharyngeal carcinoma biotherapy.
