转移抑制基因(MTSS1)、微血管密度(MVD)在鼻咽非角化性鳞癌组织中的表达及其意义

【摘要】:目的本研究采用免疫组织化学技术检测肿瘤转移抑制基因(MTSS1)及微血管密度(MVD)在鼻咽非角化性鳞癌中的表达及二者之间相关性,探讨它们在肿瘤发生、发展及其与组织学类型及分化程度、临床分期、发生淋巴结转移情况等的关系。 方法采用免疫组织化学PV-9000二步法,检测54例鼻咽非角化性鳞癌及25例鼻咽慢性炎症组织中的MTSS1、CD105标记的MVD的表达水平。 结果MTSS1蛋白在鼻咽非角化性鳞癌组织及鼻咽炎症组织中阳性表达率分别为70.37%和92.00%,,差异具有显著性,P=0.03;MTSS1蛋白的表达在不同的TNM分期、组织学分型和是否发生淋巴转移组间的阳性表达率具有显著差异,均P0.05;CD105标记的MVD在鼻咽非角化性鳞癌肿瘤周围组织组织中的微血管计数(13.74±3.47)显著高于鼻咽慢性炎症组织(9.23±3.06),P=0.00,MVD计数在不同TNM分期、组织学分型和是否发生淋巴转移组间的微血管计数具有显著差异,均P0.05;MTSS1、MVD二者在不同年龄、性别、吸烟史和原发部位的鼻咽非角化性鳞癌组别间,表达均无显著差异,P0.05;鼻咽非角化性鳞癌组织中MTSS1蛋白表达与MVD之间尚不能认为具有相关性,P0.05。 结论MTSS1蛋白在鼻咽非角化性鳞癌中可能对肿瘤发展及转移具有抑制作用,其表达降低可能与肿瘤恶性生物学行为或预后不良有关;MVD在鼻咽非角化性鳞癌组织中高表达与肿瘤具有更高的侵袭性及预后较差有关;同时,MTSS1和MVD在鼻咽非角化性鳞癌组织中表达不具有相关性,可能两者在鼻咽非角化性鳞癌的发生、转移过程中是两个相互独立因素。
[Abstract]:Objective to investigate the expression of tumor metastasis suppressor gene (MTSS1) and microvessel density (MVD) in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC) by immunohistochemical technique. To investigate their relationship with histologic type and differentiation, clinical stage, lymph node metastasis and so on. Methods Immunohistochemical PV-9000 two-step method was used to detect MTSS1 in 54 cases of nasopharyngeal non-keratinizing squamous cell carcinoma and 25 cases of nasopharyngeal chronic inflammation. The expression level of MVD labeled by CD105. Results the positive expression rates of MTSS1 protein in nasopharyngeal non-keratinized squamous cell carcinoma and nasopharyngeal inflammation were 70.37% and 92.00, respectively. There were significant differences in the expression of MTSS1 protein between different TNM stages, histological classification and lymphatic metastasis group (P 0.05). The microvessel count of CD105 labeled MVD in the tissues surrounding nasopharyngeal non-keratinizing squamous cell carcinoma (NSCC) was 13.74 卤3.47, significantly higher than that in chronic nasopharyngeal inflammation (9.23 卤3.06). . There was significant difference in MVD count between different TNM stages, histological classification and lymphatic metastasis group (P 0.05). There was no significant difference in the expression of MTSS1 and MVD between different age, sex, smoking history and primary nasopharyngeal non-keratinizing squamous cell carcinoma (NSCC) group (P 0.05). There is no correlation between the expression of MTSS1 protein and MVD in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC). Conclusion MTSS1 protein may inhibit tumor development and metastasis in nasopharyngeal non-keratinized squamous cell carcinoma, and its lower expression may be related to malignant biological behavior or poor prognosis of the tumor. The high expression of MVD in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC) was associated with higher invasion and poor prognosis. There was no correlation between MTSS1 and MVD expression in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC). The expression of MTSS1 and MVD in nasopharyngeal non-keratinized squamous cell carcinoma (NSCC) may be an independent factor in the metastasis of nasopharyngeal non-keratinized squamous cell carcinoma (NSCC).
