葡萄糖转运蛋白1对人喉鳞状细胞癌的影响的初步研究

【摘要】: 目的观察葡萄糖转运蛋白1(glucose transporter 1,GLUT1)在人喉鳞状细胞癌中的表达情况及其与临床病理学的关系;探讨GLUT1对喉癌淋巴转移的影响。 方法应用免疫组织化学的方法以及逆转录-聚合酶链反应(RT-PCR)法检测57例喉鳞状细胞癌患者的肿瘤组织、癌旁喉组织以及远离肿瘤的正常喉组织中GLUT1的蛋白和mRNA的表达,并结合喉癌患者的临床病理特征进行分析;通过免疫组化方法,用VEGFR-3标记淋巴管,分析GLUT1对喉癌淋巴转移的作用。 结果(1)GLUT1免疫组化:GLUT1在喉癌组织中的表达高于癌旁喉组织和正常喉组织(P0.01);在低分化喉癌中的表达高于高分化喉癌(P=0.043);相比Ⅰ期+Ⅱ期喉癌,Ⅲ期+Ⅳ期喉癌有高表达GLUT1的趋势(P=0.003);有淋巴转移的喉癌的表达高于无淋巴转移的喉癌(P=0.021);在直径≥3cm的肿瘤中的表达高于直径3cm的肿瘤(P=0.032)。(2)RT-PCR结果:GLUT1在喉癌组织中的表达高于癌旁喉组织和正常喉组织(P0.01);在低分化喉癌中的表达高于中分化和高分化喉癌(P=0.026);相比Ⅰ期+Ⅱ期喉癌,Ⅲ期+Ⅳ期喉癌有高表达GLUT1的趋势(P=0.018);有淋巴转移的喉癌的表达高于无淋巴转移的喉癌(P=0.04);在直径≥3 cm的肿瘤中的表达高于直径3 cm的肿瘤(P=0.048)。(3)VEGFR-3免疫组化:在喉癌组织中的表达高于癌旁喉组织和正常喉组织(P0.01);有淋巴转移的喉癌组织的表达高于无淋巴转移的喉癌(P=0.012)。 结论GLUT1与喉癌发生、发展、分化、淋巴转移、转归等密切相关,可以作为喉癌的诊断、治疗、预后指标之一。
[Abstract]:Objective to investigate the expression of glucose transporter 1glucose transporter 1 (GLUT1) in human laryngeal squamous cell carcinoma (LSCC) and its relationship with clinicopathology. To investigate the effect of GLUT1 on lymphatic metastasis of laryngeal carcinoma. Methods the tumor tissues of 57 patients with laryngeal squamous cell carcinoma were detected by immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR). The expression of GLUT1 protein and mRNA in paracancerous laryngeal tissues and normal laryngeal tissues away from the tumor was analyzed in combination with the clinicopathological features of laryngeal cancer patients. The role of GLUT1 in lymphatic metastasis of laryngeal carcinoma was analyzed by immunohistochemical method and VEGFR-3 labeled lymphatic vessels. Results Immunohistochemical staining of 1% GLUT1 in laryngeal carcinoma was higher than that in adjacent laryngeal tissue and normal laryngeal tissue (P0.01).
