鼻咽癌组织中EB病毒编码的LMP-1与Ets-1和CyclinD1三蛋白之间的相关性研究

【摘要】： 目的：诸多研究表明,EB病毒膜蛋白LMP-1在鼻咽癌的发生发展中起重要作用,然而其具体发病机制远未阐明。我们前期研究发现：在鼻咽癌中,LMP-1与Cyclin D1的表达呈正相关。令人关注的是,新近研究发现LMP-1可以激活信号转导通路Ras/MAPKKK/MAPKK/ERK/Ets-1,激活Ets-1,同时又有研究证实,在鼠源肾细胞中,Ets-1在靶基因cyclin D1启动子上有结合位点(5'-GATCTCGAGCAGGAAGTTC GA-3'),可以激活cyclin D1的启动子,上调cyclin D1的表达。因此,我们推测在鼻咽癌中LMP-1、Ets-1和Cyclin D1三者之间存在关联。本项目拟从临床组织水平探讨LMP-1、Ets-1和Cyclin D1蛋白在鼻咽癌中的相关性及它们的表达,为进一步研究LMP-1介导的致病机制提供实验依据。方法：我们采用免疫组织化学Envision法在鼻咽癌(非角化鳞状细胞癌)活检蜡块标本中检测LMP-1、Ets-1和Cyclin D1的表达并分析它们之间的相关性,其中男性96例,女性47例,平均年龄48.2±14.8岁,年龄范围16至82岁。结果： ●LMP-1、Ets-1和Cyclin D1蛋白在鼻咽癌中的阳性表达率：143例鼻咽癌病理标本的免疫组化评价结果显示,LMP-1阳性表达率81.8%(117/143)；Ets-1阳性表达率95.1%(136/143)；Cyclin D1阳性表达率91.6%(136/143),三者在鼻咽癌中均高表达。 ●LMP-1、Ets-1和Cyclin D1在鼻咽癌中的表达相关性：相关性分析结果显示：LMP-1的高表达与Ets-1和Cyclin D1的表达呈正相关(LMP-1与Ets-1,P=0.000,r=0.397；LMP-1与Cyclin D1, P=0.001,r=0.315)；同时Ets-1表达与Cyclin D1的表达在统计上显著相关(P=0.004,r=0.368)。结论：在鼻咽癌组织中,LMP-1、Ets-1和Cyclin D1高表达,并且表达密切相关。
[Abstract]:Objective:. Many studies have shown that Epstein-Barr virus membrane protein LMP-1 plays an important role in the development of nasopharyngeal carcinoma, but its specific pathogenesis is far from clear. Our previous study found that the expression of LMP-1 and Cyclin D1 was positively correlated in nasopharyngeal carcinoma. Recent studies have found that LMP-1 can activate the signal transduction pathway Rasr / MAPKK / MAPKK / ERK- 1, activate Ets-1. At the same time, it has been confirmed that Ets-1 has a binding site on the target gene cyclin D1 promoter on the target gene cyclin D1 promoter, which can activate the promoter of cyclin and up-regulate the expression of cyclin D1. We speculated that there was a correlation between LMP-1Ets-1 and Cyclin D1 in nasopharyngeal carcinoma. This study aims to investigate the relationship between LMP-1Ets-1 and Cyclin D1 proteins and their expression in nasopharyngeal carcinoma at the clinical tissue level. To provide experimental basis for further study of LMP-1 mediated pathogenesis. Methods:. Immunohistochemical Envision method was used to detect the expression of LMP-1Ets-1 and Cyclin D1 in paraffin specimens of nasopharyngeal carcinoma (non-keratinized squamous cell carcinoma). The correlation between LMP-1Ets-1 and Cyclin D1 was analyzed in 96 males and 47 females with an average age of 48.2 卤14.8 years. The age range is 16 to 82 years. Results:. The positive expression rate of LMP-1Ets-1 and Cyclin D1 protein in nasopharyngeal carcinoma was evaluated by immunohistochemistry in 143 cases of nasopharyngeal carcinoma. The results showed that the positive expression rate of LMP-1 was 81.8%. The positive rate of Ets-1 was 95.1% 136 / 143Cyclin D1, and all of them were highly expressed in nasopharyngeal carcinoma (NPC). Correlation between the expression of LMP-1Ets-1 and Cyclin D1 in nasopharyngeal carcinoma: the results of correlation analysis showed that the high expression of LMP-1 was positively correlated with the expression of Ets-1 and Cyclin D1, and the expression of LMP-1 was positively correlated with that of Ets-1 and Cyclin D1. The expression of LMP-1 was positively correlated with the expression of Cyclin D1, and the expression of Ets-1 was significantly correlated with the expression of Cyclin D1. Conclusion:. The expression of LMP-1Ets-1 and Cyclin D1 was highly expressed in nasopharyngeal carcinoma (NPC), and the expression was closely related to the expression of LMP-1Ets-1 and Cyclin D1.
