变应性鼻炎外周血Th1、Th2及Th17细胞功能研究

【摘要】:目的:探讨常年变应性鼻炎患者外周血中Th17细胞的功能状况。方法:选取尘螨过敏性常年性变应性鼻炎患者(AR)及健康对照者(HC)各10人。从外周血分离单个核细胞(PBMCs),分为离子霉素组(空白组),尘螨组, PMA组:离子霉素组加入离子霉素1ug/ml和莫能霉素2umol/l;尘螨组加入离子霉素1ug/ml和尘螨变应原nDer p1 1ug/ml及莫能霉素2umol /l;PMA组加入PMA 20ng/ml,离子霉素1ug/ml和莫能霉素2umol/l,在37°C,5%CO2细胞培养箱培养6h。用流式细胞技术检测Th17细胞占CD4+T细胞百分比,分析常年变应性鼻炎患者外周血中Th17细胞的功能。结果:HC空白组、尘螨组和PMA组Th17细胞百分比均低于对应的AR(p 0.05);经PMA刺激后,HC和ARTh17细胞百分比较空白组均增高(p 0.05), AR增幅更大;经nDerp1刺激后,HCTh17百分比较空白组无明显改变,而ARTh17百分比较空白组显著降低(p 0.05)。结论:提示Th17细胞可能参与变应性鼻炎的发病机制。PMA刺激可以引起能分泌IL-17的CD4+T数量增加。 目的:探讨常年变应性鼻炎患者外周血中记忆性Th1和Th2细胞的功能状况。方法:选取尘螨过敏性常年性变应性鼻炎患者(AR)及健康对照者(HC)各10人。从外周血分离单个核细胞,分为离子霉素组(空白组),尘螨组,PMA组:离子霉素组加入离子霉素1ug/ml和莫能霉素2umol/l;尘螨组加入离子霉素1ug/ml和尘螨变应原nDer p1 1ug/ml及莫能霉素2umol /l;PMA组加入PMA 20ng/ml,离子霉素1ug/ml和莫能霉素2umol/l,在37°C,5%CO2细胞培养箱培养6h。培养6小时后,收集PBMCs,进行胞膜和胞内染色,用流式细胞技术检测记忆性Th1/Th2细胞占CD4+CD45RO+T(Tm)细胞百分比,分析常年变应性鼻炎患者外周血中记忆性Th1/Th2细胞的功能。结果:HC空白组外周血中记忆性的Th2百分比高于AR空白组,经尘螨刺激后,HC记忆性的Th2百分比无明显改变,而AR增加,且效应性的Th2百分比增加更明显(p 0.05)。HC外周血中记忆性的Th1百分比高于AR,经尘螨刺激后,HC和AR均下降,但是HC下降更明显,虽然没有统计学差异(P0.05)。经PMA刺激后,HC和AR外周血中记忆性的Th2百分比均显著下降(p 0.05);HC和AR外周血中记忆性的Th1百分比均显著上升(p 0.05)。结论:AR存在对尘螨变应原的特异性免疫记忆性Th2细胞。PMA可以使CD4+T细胞分泌IFN -γ增加,分泌IL-4减少。
[Abstract]:Objective: to investigate the function of Th17 cells in peripheral blood of patients with perennial allergic rhinitis. Methods: 10 patients with allergic perennial allergic rhinitis and 10 healthy controls were selected. Mononuclear cells were isolated from peripheral blood. PBMCs were divided into ionomycin group (blank group, dust mite group, PMA group: ionomycin 1ugrml and monomycin 2umolrl added in ionomycin group; dust mites group added ionomycin 1ugrml and dust mite allergen nDer p11ugrml and monenomycin 2umoll; PMA group added PMA 20ng.ml. The ratio of Th17 cells to CD4 T cells was detected by flow cytometry, which was cultured in 37 掳C ~ (5) CO _ 2 cell incubator at 37 掳C ~ (5) for 6 h. The function of Th17 cells in peripheral blood of patients with perennial allergic rhinitis was analyzed. Results the percentage of Th17 cells in the control group, dust mite group and PMA group was lower than that in the corresponding AR(p 0.05% group, and the percentage of Th17 and ARTh17 cells after PMA stimulation was higher than that in the control group. After nDerp1 stimulation, HCTh17% did not change significantly compared with the control group, while the ARTh17 percentage decreased significantly compared with the control group (P 0.05). Conclusion: the results suggest that Th17 cells may participate in the pathogenesis of allergic rhinitis. PMA stimulation can induce the increase of CD4 T secreting IL-17. Methods: ten patients with allergic perennial allergic rhinitis and 10 healthy controls were selected to isolate mononuclear cells from peripheral blood. The function of memory Th1/Th2 cells in peripheral blood of patients with perennial allergic rhinitis was analyzed. Results the percentage of Th2 in peripheral blood of the control group was higher than that of the group of AR blank, but the percentage of Th2 of memory of HC was not changed after stimulation by dust mites, but AR increased. The percentage of effectual Th2 in peripheral blood was significantly higher than that of Th1 in peripheral blood, but HC and AR decreased significantly after dust mites stimulation, but the decrease of HC was more obvious. Although there was no statistical difference (P 0.05), the percentage of memory Th2 in peripheral blood of HC and AR decreased significantly after stimulation by PMA. The percentage of Th1 of memory in both HC and AR increased significantly. Conclusion\% AR has a positive effect on dust mite allergen. Specific immune memory Th2 cells. PMA could increase the secretion of IFN-纬 by CD4 T cells. Secretion of IL-4 decreased.
