膜联蛋白A5在喉癌组织中的表达及其临床意义研究

【摘要】：目的：研究膜联蛋白A5（annexinA5，ANXA5）在喉癌组织中的表达情况，探讨膜联蛋白A5在喉癌发生、发展过程中的作用及其临床意义。 方法：采集川北医学院附属医院耳鼻咽喉科手术切除的喉癌组织53例和癌旁组织25例，所有标本均一式三份：一份送病检，一份制作成石蜡标本保存，一份置液氮罐编号保存备用。所有喉癌标本均经病理学诊断为喉鳞状细胞癌。采用逆转录聚合酶链反应技术（RT-PCR）从RNA水平检测annexin A5mRNA在喉癌组织和癌旁组织中的表达水平；运用免疫组织化学染色技术链霉菌抗生物素蛋白-过氧化物酶连接法（SP法）从蛋白质水平检测annexin A5蛋白在喉癌组织和癌旁组织中的表达情况。 结果：喉癌与癌旁正常组织中ANX A5的mRNA存在表达差异，annexin A5mRNA/β-action分别为0.57987±0.98881和0.45675±0.10801，差异比较具有统计学意义（P0.05）。喉癌和癌旁正常组织中annexin A5的阳性高表达率分别为69.81％（37／53）和32.00％（8／25），比较两者差异具有统计学意义（P0.05）；在Ⅰ-Ⅱ期和Ⅲ-Ⅳ期喉癌组织中annexin A5的阳性高表达率分别为42.86％（9／21）和87.50％（28／32），在病理学分级方面，，高分化、中分化和低分化喉癌组织中annexin A5的阳性高表达率分别为37.04％（10／27）、63.16％（12／19）和85.71％（6／7），同时annexin A5在伴有淋巴结转移和不伴有淋巴结转移的喉癌组织中的阳性高表达率分别为88.24％（15／17）和52.78％（19／36），它们之间的差异均具有统计学意义（P0.05）。而annexin A5在不同年龄、性别及临床分型中的阳性高表达率分别为年龄≥55岁：65.00％（26／40）、＜55岁：84.62％（11／13）；男性：58.33％（28／48）、女性：80.00％（4／5）；声门上型：62.50％（10/16）、声门型：60.00％（18／30）、声门下型：57.14％（4／7）；差异比较均无统计学意义（P0.05）。 结论：喉癌组织中annexin A5的mRNA与蛋白质都存在高表达；annexin A5蛋白的表达水平与患者临床分期、病理学分级和颈淋巴结转移密切相关，与患者年龄、性别及临床分型无关。因此，annexin A5可能在喉癌的发生、发展中发挥重要作用。
[Abstract]:Objective: to study the expression of annexin A 5 (A5) in laryngeal carcinoma, and to explore the role of A5 in the carcinogenesis and development of laryngeal carcinoma and its clinical significance. Methods: 53 cases of laryngeal carcinoma and 25 cases of adjacent tissues were collected from Otolaryngology Department of affiliated Hospital of North Sichuan Medical College. All laryngeal carcinoma specimens were pathologically diagnosed as laryngeal squamous cell carcinoma. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of annexin A 5 mRNA in laryngeal carcinoma and adjacent tissues. Immunohistochemical staining technique was used to detect the expression of annexin A5 protein in laryngeal carcinoma and paracancerous tissues. Results: the differences of ANX A5 mRNA expression in laryngeal carcinoma and adjacent normal tissues were 0.57987 卤0.98881 and 0.45675 卤0.10801, respectively.
