贝伐单抗对人脐静脉内皮细胞纤维化相关炎症因子表达的影响

【摘要】:目的观察贝伐单抗对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC)纤维化相关炎症因子表达的影响,以探讨贝伐单抗治疗后新生血管纤维化的可能机制。方法分别在正常氧含量及缺氧条件下培养HUVEC,培养液中加入终浓度为0.25g·L-1贝伐单抗,根据处理时间的不同分为0 h组(对照组)、6 h组、12 h组、24 h组、48 h组。分别用RT-PCR及ELISA方法检测各组纤维化相关炎症因子白细胞介素(interleukin,IL)-1β、IL-6、IL-8以及肿瘤坏死因子-α(tumor necrosis factor-alpha,TNF-α)mRNA和蛋白的表达。结果正常氧含量条件下,与对照组相比,IL-6、IL-8的mRNA和蛋白表达在各实验组均明显增加,而IL-1β、TNF-αmRNA和蛋白的表达仅在12 h、24 h、48 h组明显增加,差异均有统计学意义(均为P0.05)。缺氧条件下,与对照组相比各实验组IL-1β、IL-6、IL-8及TNF-α的mRNA和蛋白表达在各时间点均明显增加,差异均有统计学意义(均为P0.05)。结论无论在正常氧含量还是缺氧条件下,贝伐单抗均可刺激纤维化相关炎症因子的表达增加,此部分炎症因子可能参与了抗新生血管内皮生长因子治疗后的新生血管纤维化过程。
[Abstract]:Objective to observe the effect of bevacizumab on the expression of inflammatory cytokines associated with fibrosis in human umbilical vein endothelial cells (HUVECs) of human umbilical vein endothelial cells (HUVECs). Methods HUVECs were cultured under the condition of normal oxygen content and hypoxia respectively, and the final concentration of bevacizumab was 0.25 g 路L ~ (-1). According to the time of treatment, they were divided into 0 h group (control group, 6 h group, 12 h group, 24 h group, 48 h group, respectively). RT-PCR and ELISA methods were used to detect interleukin-1 尾 -IL-6 IL-8, tumor necrosis factor- 伪 tumor necrosis factor- 伪 mRNA and TNF- 伪 mRNA, respectively. Results under the condition of normal oxygen content, Compared with the control group, the expression of mRNA and protein of IL-8 was significantly increased in all experimental groups, while the expression of TNF- 伪 mRNA and protein in IL-1 尾 -TNF- 伪 increased only at 12 h and 24 h after 48 h, the difference was statistically significant (all P0.050.The expression of TNF- 伪 mRNA and protein in hypoxia group was significantly higher than that in control group. Compared with the control group, the expression of mRNA and protein of IL-1 尾 -IL-6, IL-8 and TNF- 伪 in each experimental group were significantly increased at each time point, and the difference was statistically significant (P 0.05). Conclusion the expression of IL-8 and TNF- 伪 in the experimental group is significantly higher than that in the control group (P 0.05). Bevacizumab can stimulate the expression of fibrosis-related inflammatory factors, which may be involved in the process of neovascularization after anti-angiogenic endothelial growth factor treatment.
